摘要
目的 采用RP -HPLC测定 5 -氨基水杨酸 (5 -ASA)壳聚糖胶囊的含量。方法 衍生化使样品中 5 -ASA乙酰化 ,色谱柱为Nucleosil1 0 0 - 5C1 8,流动相为磷酸盐缓冲液 -乙腈 -四氢呋喃 (85∶1 0∶5 ) ,流速 0 .9ml·min-1 ,检测波长 2 5 4nm。结果 5 -ASA在 0 .78~ 5 0 μg·ml-1 范围内线性关系良好 (r=0 .9993) ,RSD =1 .2 6 % (n =5 )。结论 所用方法可测定 5
OBJECTIVE To establish an RP-HPLC method to determine the 5-aminosalicylic acid (5-ASA) in Chitosan capsule.METHODS 5-ASA was derivatized to Acetyl 5-ASA.RP-HPLC was adopted with a Nucleosil 100-5 C 18 column, a mobile phase consisting of phosphate buffer-acetonitrile-tetrahydrofuran (85∶10∶5),at 0.9 ml·min -1 . The UV detective wavelength was 254 nm.RESULTS 5-ASA was well separated, the method showed a good linearity for 5-ASA in the range of 0.78-50 μg·ml -1 ( r =0.9993), the lowest limit of detection was 0.04 μg·ml -1 .CONCLUSION The method can be used to determine 5-ASA.
