以三碘甲腺原氨酸衍生物(T_3X)和Na^(125)I为原料,用氯胺-T法合成了^(125)I-T_3X和^(125)I-T_4X经HPLC分离纯化,产品的比活度:^(125)I-T_3X为16.87—31.78GBq/mg,^(125)I-T_4X为31.15—46.07GBq/mg。该二种示踪剂已用于直接法放射免疫分析测定血中游离T_3和游离T_4浓度,并已证明,该衍生物虽经化学改性限制了与血清中内源蛋白的结合.但仍保持了T_3、T_4原有的与相应特异性抗体的结合特性。
Using triiodothyronine derivative(T3X) and Na 125I as starting materials, 125I-T3X and 125I-T4X were synthesized by the chloramine-T method and separated by HPLC. The specific activity of products was 16.87-31.78GBq/mg for 125I-T3X and 3] 15-46.07 GBq/mg for 125I-T4X. The tracers have been used in the direct method of RIA for free T3 and free T4 in serum. It's proved that the derivative, chemically modified to inhibit its binding to the endogenous serum proteins, still remain original binding characteristics of T3, T4 with the specific antiserum.
