摘要
应用马来酸酐和苯乙烯聚合成共聚物,并将它配成丁酮-乙腈溶液,把共聚物固定到聚苯乙烯试管内壁上,借此将二抗IgG结合到试管壁上,制备成二抗固化管.利用该管建立了固相放免分析血浆ACTH以及双抗固相分离血清皮质醇RIA.所建方法的标准曲线、相关性、精密度、准确性及灵敏度都十分满意,与各自液相方法对照,测定病人样品,结果一致,两者相关密切.这些固相方法简化了操作步骤,便于推广应用.
Abstract It is difficult to immobilize antibody by the formation of covalent bonds on polystyrene tubes. As maleic anhydride-styrene copolymer is able to bind NH2groups of proteins, the copolymer was synthesized with maleic anhydride and styrene, and dissolved in solution of methylethyl ketone-acetonitrile. The polystyrene tubes (12mm × 75mm) were coated with copolymer solution and made into“activated tubes”. Aliquot of 0.5ml of the second antibody (AB2) IgG (about 300 ̄500μg in 0.1M citrate buffer, PH7.5) was added into each activated tube and incubated for 16 hours at room temperature, so that 30  ̄ 40μg IgG was immobilized on the surface of each activated tube, which was referred to as “AB2-immobilized tube”. Two kinds of solid phase assay have been developed with AB2-immobilized tubes. The reproducibilities of intra and interassay of these solid phase assays were quite satisfactory. Their analytical recoveries falled in the range from 94.9% to 114.3%.
