摘要
目的 :建立去甲万古霉素血药浓度的快速高效液相色谱测定法。方法 :血样处理采用 Zn SO4 沉淀法 ,色谱分析采用 Diam onsil TM(钻石 ) C1 8色谱分析柱 ,流动相为甲醇 -磷酸二氢钾缓冲液 (p H 3.2 ) =2 4∶ 76 ,紫外检测波长 2 36 nm ,灵敏度为0 .0 15 AUFs,实验数据用 Gilson715液相色谱工作站处理 ,以峰面积、外标法定量。结果 :本法在万古霉素浓度 (x)为 0 .5~6 0 μg/ ml间呈良好线性关系 ,相关系数 r=0 .9998(n=6 ) ,血浆最低检测浓度为 0 .2 μg/ m l(S/ N≥ 2 ) ,日内、日间 RSD均 <6 %。结论 :此法采血量少、快速简便、灵敏度和准确度高 ,适合临床常规血药浓度监测及实验课题研究。
Objective: To establish a rapid RP-HPLC method for determination of norvancomycin plasma level. Methods: The pretreatment of the blood samples was simply sedimentation with 10% ZnSO 4. Then the sample was separated on Diamonsil TM C 18 column with a mixture of methanol and KH 2PO 4 buffer solution (24∶76) as the mobile phase. The UV detector wavelength was set to 236 nm and sensitivity was 0.015 AUFS. The experiment data were treated with Gilson 715 chromatographic workstation and calculated by peak area and external standard method. Results: A linearity was obtained within the range of norvancomycin concentrations from 0.5-60 μg/ml ( r=0.999 8, n =6), the inspect limitation was 0.2 μg/ml (S/N≥2), and the intra-day and inter-day RSD were less than 6%. Conclusion: It appears to be a rapid, convenient and accurate method for determination of the norvancomycin concentrations in plasma.
