简单灵敏的高效液相色谱法测定人血浆中去甲万古霉素的浓度

摘要 目的建立血浆中去甲万古霉素含量的HPLC测定方法。方法采用高效液相色谱检测法,分析柱为Tskgel ODS-80TS(4.6mm×250mm,5μm),流动相为10mmol·L^-1甲酸铵(0.1%甲酸)-甲醇(0.1%甲酸)=(82:18,v/v),流速:1.0mL·min^-1,柱温:30℃,检测波长:236nm,进样量:20μL.血浆样品用三氟醋酸直接沉淀后,进样分析。结果去甲万古霉素在1~1000μg·mL^-1与峰面积线性关系良好(r=0.999),最低检测浓度为1μg·mL^-1。平均回收率为96%;日间、日内RSD均〈15%。结论研究建立的HPLC法快速、简便、高效,精密度和准确度高,适合血样中去甲万古霉素浓度的检测。 Objective To determine the content of norvancomycin in human plasma by HPLC. Methods High-performance liquid chromatography with UV detection was used. Tskgel ODS-80TS (4.6 mm×250mm,5μm) was the analysis column. The mobile phase was 10 mmol · L^-1 ammonium formate (0. 1% formic acid) -methanol (0. 1 formic acid) = (82 : 18, v/v), the flow rate was 1.0 mL·min^-1, and the column temperature was 30 ℃. The detection wavelength was 236 nm. The injection volume was 20μL. The plasma sample was directly precipitated with trifluoroacetic acid, and the transfer supernatant was analyzed after the centrifugation. Results Norvancomycin was lin ear within 1--1 000μg·mL^-1 (r=0. 999). The lowest detectable concentration was 1 μg·mL^-1. The average re covery was 96%. The RSD of intra-day or inter-day was less than 15%. Conclusion The HPLC method is rapid, simple, convienient and efficient, and suitable for the determination of norvancomvcin.
