四逆汤对供心热缺血保护作用的实验研究 被引量:1

摘要 目的 :探讨四逆汤对供心热缺血的保护作用 ,以达到延长供心耐受热缺血时限的目的。方法 :新西兰大白兔 16只随机分2组 ,每组 8只。 A组为对照组 ,B组为实验组。B组取心前连续四逆汤汤剂灌胃 3 d。热缺血 10 m in后 ,A组用 UW液灌注 ,B组用 UW液加四逆汤注射液灌注 ;取心后 ,A组在 UW液、B组在 UW+四逆汤注射液中保存 4 h。然后在 langendoff模型上A组用氧合的 KHB液、B组用 KHB液加四逆汤注射液灌注 3 0 min。以功能、代谢和结构等指标评价供心恢复情况。结果 :B组各项指标均优于 A组。结论 :用四逆汤对供心作预处理 ,并在灌洗液、保存液和再灌注液中加入适量的四逆汤 ,对供心热缺血有保护作用 。 Objective:To explor the proctective effect of SD on heart warm ischemia Methods:16 New Zealand rabbits[WT:(2 51+0 14) kg],were devided into two groups The rabbits in Group B were fed with SD at the dose of 5 g/kg·d -1 for three days before obtaining the hearts,and the rabbit's hearts in Group A and B weren't reperfused until in heat warm ischemia condition for 10 minutes Hearts donor in Group A were perfused with UW solution at 4 ℃,and Group B with UW and Sini Decoction(10 ml/L) at 4 ℃ After the hearts were taken out, preservation at 4 ℃ for 4 hours was given in Group A with UW solution, and Group B with UW+SD(10 ml/L) Then,the hearts were reperfused with KHB solution in langendorff model at 37 ℃ for 30 minutes SD was added into KHB solution in Group B The measured indexes are ①the time from the beginning of reperfusion to the recovery of stable heart beat rhythm ②the heart rate in stable heart beats ③the recoverable power of myocardial contraction in stable heart beats ④the coronary sinus flow(CSF) of heart and the myocardial oxygen consumption (MVO 2) in reperfusion at 0,5,10,15 and 30 minutes ⑤ATP , MDA and SOD content of myocardium before and at the end of reperfusion ⑥left ventricle myocardium was obtained for electron microscope at the end of reperfusion Result:There is significant difference between Group B and Group A(P<0 05) Conclusions:SD has the protective effect on warm ischemia donor heart
