射干中野鸢尾黄素对合胞病毒感染豚鼠感染后咳嗽及气道炎症的影响 被引量:8

Effects of Irigenin in Shegan(Belamcanda chinensis) on Cough and Airway Inflammation in Guinea Pigs Infected with Respiratory Syncytial Virus

摘要 目的:研究射干野鸢尾黄素对呼吸道合胞病毒(RSV)感染豚鼠咳嗽模型气道炎症的治疗作用。方法:采用烟熏配合RSV滴鼻建立豚鼠咳嗽后模型,造模结束后给予野鸢尾黄素治疗,中药阳性组给与肺宁片治疗。通过枸橼酸雾化诱咳比较各组豚鼠的咳嗽次数;采用HE染色法对肺组织形态学检测;血液体液分析仪检测肺泡灌洗液中白细胞总数及分类计数;采用ELISA试剂盒法检测肺泡灌洗液中细胞因子,采用Western Blot法检测肺组织中T-bet和GATA-3蛋白的表达。结果:豚鼠肺组织病毒培养液中显示RSV感染成功,模型组豚鼠与空白组相比,在5 min内咳嗽次数显著升高(P<0.01),肺泡灌洗液中除嗜碱性粒细胞计数外,其它各炎性细胞显著升高(P<0.05),同时IFN-γ显著降低(P<0.05),模型组IL-4含量和IL-4/IFN-γ比值升高(P<0.05),GATA-3蛋白表达水平显著升高且T-bet蛋白表达水平显著降低(P<0.05)。给药后,野鸢尾黄素组豚鼠5min内咳嗽次数和肺泡灌洗液中嗜酸性粒细胞显著下降(P<0.05),IFN-γ显著升高(P<0.05),IL-4/IFN-γ比显著下降(P<0.05),肺组织中T-bet蛋白表达水平显著升高(P<0.05)。结论:射干野鸢尾黄素对RSV感染后咳嗽模型豚鼠具有较好的镇咳作用,具有抗气道炎症作用。 Objective:To study the therapeutic effect of the irigenin on airway inflammation in the guinea pig cough model infected with respiratory syncytial virus(RSV).Methods:The model of guinea pig cough was established by using smoked and RSV nasal drops.After the model was established,the treatment with irigenin was given and the positive Chinese medicine group was given Feining Tablets.The cough frequency of each group of guinea pigs was compared by citrate atomization induced cough.The morphology of lung tissue was detected by HE staining.The total number and count of white blood cells in alveolar lavage fluid were detected by blood body fluid analyzer.Cytokines in alveolar lavage fluid were detected by ELISA kit method.The expressions of T-bet and GATA-3 protein in lung tissue were detected by Western Blot.Results:The guinea pig lung tissue virus culture medium showed successful RSV infection.Compared with the control group,the number of coughs in the model group was significantly increased within 5 min(P<0.01).In addition to the basophil count,the other inflammatory cells were significantly elevated in the alveolar lavage fluid(P<0.05).At the same time,IFN-γwas significantly decreased(P<0.05),and the IL-4 content and IL-4/IFN-γratio in the model group were increased(P<0.05).The expression level of GATA-3 protein was significantly increased and the expression level of T-bet protein was significantly decreased(P<0.05).After administration of irigenin,the number of coughs and eosinophils in the alveolar lavage fluid of the guinea pigs in the irigenin group decreased significantly(P<0.05).The level of IFN-γwas significantly increased(P<0.05)and the IL-4/IFN-γratio decreased significantly(P<0.05).The expression level of T-bet protein in lung tissue was significantly increased(P<0.05).Conclusion:The irigenin has a good antitussive effect on guinea pigs with cough model after RSV infection,and has anti-airway inflammation.
