摘要
目的:研究长链非编码RNA(LncRNA)PVT1在LPS诱导THP-1源巨噬细胞炎症反应中的表达及其与炎症基因表达相关性,推测lncRNA-PVT1在脓毒症急性肺损伤炎症反应中的可能作用。方法:通过LPS刺激THP-1源巨噬细胞产生炎症反应造模,利用RT-qPCR检测,比较不同刺激浓度(0、0.5、5.0μg/ml,8 h)下各组细胞中lncRNA-PVT1及TNF-α、IL-1βmRNA表达情况。结果:与对照组相比,IL-1β及TNF-αmRNA在LPS不同刺激浓度下,均表达上调,且具有统计学差异(均P<0.05)。随着刺激浓度增加,IL-1β及TNF-αmRNA表达量上升,呈浓度依赖,在LPS 5.0μg/ml作用8 h下,IL-1β及TNF-αmRNA表达量达峰值。与对照组相比,在5.0μg/ml LPS组中ln-cRNA-PVT1表达明显上调,具有统计学差异(P<0.0001)。相关性分析发现5μg/ml LPS刺激8 h后,THP-1源巨噬细胞中lncRNA-PVT1相对表达量分别与IL-1β及TNF-αmRNA相对表达水平呈正相关(分别为R^2<0.6134,P=0.0125;R^2<0.7825,P=0.0015)。结论:LncRNA-PVT1在LPS致THP-1源巨噬细胞炎症反应中高表达,且与TNF-α及IL-1βmRNA表达呈正相关,提示该lncRNA-PVT1在脓毒症急性肺损伤炎症反应中可能起重要作用。
Objective:To investigate the expression of long-chain non-coding RNA(LncRNA) PVT1 in LPS-induced inflammatory responses in THP-1 derived macrophages and its correlation with inflammatory gene expressions,and to speculate the underlying mechanism that lncRNA-PVT1 is involved in the inflammatory responses of septic acute lung injury.Methods:Inflammatory responses of THP-1 derived macrophages were induced by LPS. RT-qPCR was used to detect the expressions of lncRNA-PVT1 and IL-1β,TNF-α mRNA in different concentrations of LPS(0,0.5,5.0 μg/ml,8 h).Results:Compared with the control group,IL-1β and TNF-α mRNA were elevated at different concentrations of LPS with statistical differences(both P<0.05). With the increase of LPS concentrations,the expressions of IL-1β and TNF-α mRNA increased in a concentration-dependent manner. The expressions of IL-1β and TNF-α mRNA peaked with LPS 5.0 μg/ml at 8 h. Compared with the control group,the expression of lncRNA-PVT1 was significantly up-regulated in the 5.0 μg/mL LPS group with statistical difference(P<0.0001). Further correlation analysis showed that the relative ex-pression of lncRNA-PVT1 in THP-1 derived macrophages was positively correlated with the relative levels of IL-1β and TNF-α mRNA with 5 μg/ml LPS for 8 h(R^2<0.6134,P=0.0125;R^2<0.7825,P=0.0015).Conclusion:LncRNA-PVT1 was highly expressed in LPS-induced inflammatory responses in THP-1 derived macrophages and positively correlated with TNF-α and IL-1β mRNA expressions,suggesting that lncRNA-PVT1 might exert an important role in inflammatory responses in septic acute lung injury.
