摘要
目的:比较氯己定与氟康唑、伊曲康唑、克霉唑联合体外抗念珠菌生物膜效应。方法:利用改良96孔板构建念珠菌生物膜,并参考美国临床实验室标准化研究所(clinical laboratory standard institute,CLSI)M27-A3方案及棋盘法检测氯己定与氟康唑、伊曲康唑和克霉唑对念珠菌生物膜的联合抗菌效应。结果:氯己定、氟康唑、伊曲康唑、克霉唑对念珠菌生物膜的80%最低抑菌浓度SMIC80均>32μg/mL。读取SMIC50时,氯己定与3种抗真菌药物的协同率为100%。读取SMIC80时,氯己定与3种抗真菌药物的协同率均≥80%。结论:在体外,氯己定与3种抗真菌药物联合对念珠菌生物膜时,能够降低3种药物对念珠菌生物膜的SMIC值,增强的抗念珠菌生物膜的作用。
Objective:To investigate the in vitro antifungal effects of the combinations of chlorhexidine with fluconazole,itraconazole and clotrimazole.Methods:Candida biofilms were prepared by using a modified 96-wells plates based method.The in vitro antifungal activity of the combinations of chlorhexidine with fluconazole,itraconazole and clotrimazole against Candida biofilms were investigated via the broth microdilution of M27-A2 and checkerboard technique.Results:The sessile minimum inhibitory concentration ranges resulting in 80%of Candida biofilms were all more than 32μg/mL.When the FICIs calculated by SMIC50,the synergistic effects were 100%in all combination groups.When the FICIs calculated by SMIC80,the synergistic effects were at least 80%in all combination groups.Conclusion:The combinations of chlorhexidine with fluconazole,itraconazole and clotrimazole can enhance the anti-biofilm effects against Candida biofilms.
