摘要
采用聚丙烯酰胺凝胶电泳法对成年白香猪的肺、肝、脾、肾和胰脏组织的乳酸脱氢酶(LDH)进行分离,结果显示:它们均含有5条LDH同功酶谱带。经生物图象处理系统扫描定量得出:LDH1,LDH2,LDH3,LDH4和LDH5各组分在肺脏的百分含量的排列顺序:LDH5>LDH2>LDH1>LDH3>LDH4,M亚基∶H亚基为62.93%∶37.08%;在肝脏中百分含量的排列顺序为:LDH5>LDH2>LDH1>LDH3>LDH4,M亚基∶H亚基为49.52%∶50.48%;在脾脏中百分含量排列依次是:LDH5>LDH2>LDH4>LDH3>LDH1,M亚基∶H亚基为52.41%∶47.59%;在肾脏中百分含量多少的次序是:LDH1>LDH2>LDH4>LDH3>LDH5,M亚基∶H亚基为19.00%∶81.00%;在胰脏中百分含量顺序是:LDH1>LDH2>LDH5>LDH3>LDH4,M亚基∶H亚基为32.86%∶67.14%。以上结果提示:LDH同功酶在不同的内脏组织中各谱带及亚基组成与含量的不同,它们反映了相应组织的中乳酸和丙酮酸间转化代谢过程的差异,由此保证了机体糖代谢中LDH所催化反应的正常协调进行。
Lactic dehydrogenase (LDH) isoenzyme of the lung, liver, spleen, kidney and pancreas was determined with PAGE (polyacrylamide gel electrophoresis) in adult (7-month-old) white xiang mini-pigs. Five LDH bands were detected in all the organs studied. The order of various LGH components was LDH5> LDH2> LDH1> LDH3> LDH4 in the lung, LDH5> LDH2> LDH1> LDH3> LDH4 in the liver, LDH5> LDH2> LDH1> LDH3> LDH4 in the spleen, LDH1> LDH2> LDH4> LDH3> LDH5 in the kidney and LDH1> LDH2> LDH5> LDH3> LDH4 in the pancreas. The ratio of subunit M to subunit H was 62.93%∶37.08% in the lung, 49.52%∶50.48% in the liver, 49.52%∶50.48% in the spleen, 19.00%∶81.00% in the kidney and 32.86%∶67.14% in the pancreas. The above results suggest that LDH isoenzyme and its subunit components vary with organs, which reflects the diversity of metabolic characteristics in different muscle tissues so as to ensure a coordinate carbohydrate metabolism in mini-pigs.
