摘要
目的对博落回[Macleaya cordata(Willd.)R.Br.]中苯并菲啶类生物碱外消旋体进行分离鉴定。方法采用硅胶柱色谱、氧化铝、ODS、Sephadex LH-20柱色谱和制备高效液相色谱、薄层色谱、手性液相色谱等方法进行分离纯化,通过1D NMR、2D NMR、UV、IR、X-单晶衍射及计算ECD等方法对化合物进行结构鉴定。结果从博落回乙醇提取物中分离得到11个化合物,分别鉴定为:(+)-博落回新碱A(1),(–)-博落回新碱A(2),(–)-博落回新碱C(3),(+)-博落回新碱C(4),(–)-6-丙酮基二氢血根碱(5),(+)-6-丙酮基二氢血根碱(6),(–)-6-丙酮基二氢白屈菜赤碱(7),(+)-6-丙酮基二氢白屈菜赤碱(8),(±)-6-甲氧基二氢血根碱(9),(±)-6-甲氧基二氢白屈菜赤碱(10),(±)-spallidamine(11)。结论化合物1~4为新化合物。
Objective To study the racemic benzophenanthridine alkaloids from Macleaya cordata. Methods Chromatographic methods were used for the isolation and purification,including opening silica gel,aluminium oxide,ODS,Sephadex LH-20 column chromatography,preparative TLC,semi-preparative HPLC,and chiral HPLC separation. Their structures were elucidated on the basis of 1D,2D NMR,UV,IRspectroscopic analyses,single-crystal X-ray diffraction,and electronic circular dichroism calculation. Results Eleven compounds were isolated from the ethanol extract of Macleayacordata.Their structures were identified as( +)-macleayin A( 1),( –)-macleayin A( 2),( +)-macleayin B( 3),( –)-macleayinB( 4),( –)-6-acetonyldihydrosanguinarine( 5),( +)-6-acetonyldihydrosanguinarine( 6),( –)-6-acetonyldihydrochelerythrine( 7),( +)-6-acetonyldihydrochelerythrine( 8),( ±)-6-methoxyldihydrosanguinarine( 9),( ±)-6-methoxyldihydrochelerythrine( 10),and( ±)-spallidamine( 11). Conclusion Compounds 1 ~ 4were new compounds.
