摘要
目的探讨自由基引发剂对β-环糊精(β-CD)毛细管整体柱手性药物拆分能力的影响。方法采用水溶性引发剂2,2'-偶氮二异丁腈脒二盐酸盐(AIBA)及常用的脂溶性引发剂偶氮二异丁腈(AIBN),分别制备了2根相近的环糊精整体柱poly(GMA-EDA-β-CD-co-MBA),并系统比较两者的化学性质(差示扫描量热分析和元素分析)、物理性质(通透性、重复性和扫描电镜)及色谱性能(保留机制和手性拆分评估)的差异。结果亲水性更强的AIBA能加速自由基聚合反应,但对β-环糊精整体柱的手性拆分效果并不优于AIBN。结论引发剂对手性拆分能力的影响明显,油溶性的AIBN在环糊精聚合物整体柱制备上更具优势。
Objective To investigate the effect of radical initiator on the enantioseparation ability of β-cyclodextrin( β-CD) functionalized monolithic capillary column. Methods Two poly( GMA-EDA-β-CD-co-MBA) monolithic columns have been prepared by using hydrosoluble initiator 2,2'-azobis( 2-methylpropionamidine) dihydrochloride( AIBA) and hydrophobic initiator 2,2'-azobis( 2-methylpropionitrile)( AIBN),respectively. Their differences of chemical properties( DSC analysis and elemental analysis),physical properties( permeability,reproducibility and SEM) and chromatographic properties( retention mechanism and enantioseparation evaluation) were also systematically compared. Results The results demonstrated that the more hydrophilic initiator AIBA could speed up the free radical re action,but no advantage on the enantioseparation ability of its β-CD-functionalized monolithic column was observed.Conclusion Initiator has an obvious effect on enantioseparation ability,and hydrophobic initiator AIBN has obvious advantages in the preparation of cyclodextrin polymer monolithic column.
