摘要
目的:发展一种"一步法"制备环糊精有机聚合物毛细管整体柱的方法.方法:以含单-6-甲基丙烯酰-乙二胺-6-脱氧-β-环糊精为单体,EDMA为交联剂,采用异丙醇和二甲基亚砜为生孔剂,AIBN为引发剂,通过原位聚合反应制备了poly(MAC-EDA-BCD-co-EDMA)(Column B)整体柱.经过对制备条件进行优化,获得最优比例整体柱,并对其理化性质以及手性拆分效果进行考察.最后,结合分子对接手段考察其分子识别机理.结果:与由单-6-(1H-1,2,3-三氮唑-4-甲基丙烯酸甲酯)-β-环糊精为单体制备而成的Column A相比,Column B具有更好的手性拆分效果.结论:研究表明,一步法具有良好的实际应用;Column B因为单体柔性的间隔臂连接方式,保证了环糊精空腔具备足够的柔性,故具有良好的手性分离能力.
Objective: To develop a single-step process for the preparation of β-cyclodextrin( BCD)polymeric monolithic columns. Methods: A BCD-functional capillary monolithic column poly( MACEDA-BCD-co-EDMA)( Column B) was prepared using mono-6-methacryloyl-ethylenediamine-6-deoxy-β-CD( MAC-EDA-BCD) as monomer,EDMA as crosslinker,isopropanol and DMSO as porogens and AIBN as the polymerization initiator. After the optimization of preparation conditions, a series of characterizations were conducted by physicochemical properties and chiral separation effect. Finally the mechanism of molecular reorganization was studied using Surflex-Dock. Results: Compared with Column A prepared by PMA-BCD, Column B exhibited higher enantioresolution for chiral compounds.Conclusion: It demonstrated that single-step strategy has good practical application on preparation of organic polymeric monolithic column. The satisfactory enantioselectivity may be related to the linker of the BCD monomer, which might be long enough to afford enough flexibility for efficient chiralseparations.
