摘要
通过农杆菌介导法 ,将深黄被孢霉△6-脂肪酸脱氢酶基因导入到栽培大豆吉育 4 3、黑农36、黑农 37等品种。采用多种外植体和感染方法 ,从子叶节和胚轴诱导出丛生芽与再生植株。经过在含 5 0mg L卡那霉素的筛选培养基中连续筛选 ,获得一批转基因植株。经PCR检测和DNA分子杂交分析 ,初步证明外源基因已导入并整合到大豆基因组中。
The △ 6-fatty acid desaturase gene fron Mortitieralla isabellina was transferred into cultivars Jiyu 43,Heinong 36,Heinong 37, et al. via Agrobacterium infection by inducting with various explant methods. Adentitious buds and regeneration plant were inducted from cotylendon node, hypocotyl. Kanamycin resistant (Kmr) plantlets were obtained by 3-4 cycles screening in selective condition. PCR and Southern blot analysis from the transferred gene plants proved that foreign gene were transferred and integrated into soybean genome.
