摘要
目的 :探讨在体外培养中成纤维细胞生长因子(FGF)对大鼠骨髓基质细胞诱导成骨的影响。方法 :用IMDM培养基分离、培养出大鼠胫、股骨单层贴壁的骨髓基质细胞 ;用地塞米松 (Dex)、—甘油磷酸钠、抗坏血栓 (VitC)联合诱导培养 ;然后分 :A组 ,单纯诱导 ;B~E组 ,分别加入不同浓度的FGF对骨髓基质细胞 (MSC)的诱导分化过程干涉 ;选择 2 ,8,14 ,2 0 ,2 6d时间点比较各组的增殖指数、碱性磷酸酶活性、骨钙素含量及钙化结节形成率。结果 :当加入浓度为2 ,4 ,6 ,8mg·L-1的FGF后 ,培养细胞的碱性磷酸酶活性依次降低 ,分别为 :(82± 5 ,72± 5 ,5 8± 4 ,4 8± 4 )nkat·L-1,且 4组数据间有明显差异性 (P <0 0 5 ) ,但钙化结节形成率及骨钙素含量依次增加 ,分别为 :(1 998± 0 2 2 3,2 5 79± 0 2 38,3 0 6 7± 0 2 5 9,3 70 1± 0 2 93)ng·L-1,且 4组数据间有明显差异性 (P <0 0 5 ) .结论 :FGF浓度为 2mg·L-1时 ,ALP活性最高 ,钙化结节形成速度及骨钙素含量最小 ,且三者有明显剂量依赖性 (p <0 0 5 ) .
AIM: To explore the effects of fibr ob last growth factor (FGF) on the induced osteoblast of rat bone marrow-derived m esenchymal stem cells. METHODS: The osteoblasts were induc ed from and expanded in cell culture with dexamedasone, sodium-glycerophosphate and ascorbic acid (Vit C) respectively. The induced cells with different concent rations of FGF (2, 4, 6 and 8 mg·L -1 ) as experimental group and the cells without FGF as control group were compared. The growth curve, activity of alkal ine phosphates (ALP) and osteocalcin (OCN) were observed. RESULTS: After adding FGF with different densities (2, 4, 6 and 8 mg·L -1 ), the growth rate of the cultured cells had no great differentiation ( P>0 05). The secretion of ALP decreased from 4 90±0 31 U·L -1 to 2 91±0 21 U·L -1 (P<0 05). But the osteocalcin (OCN) secretion was increased from 1 998±0 223 ng·L -1 to 3 701±0 293 ng·L -1 (P<0 05). The formation of mineralized node became increased (P<0 05). CO NCLUSION: FGF has significant effects on the inducement of Wistar rat bo ne marrow-derived mesenchymal stem cells in vitro. With the conc entration of FGF 2 mg·L -1 , ALP and OCN get to the highest in a dose-depen dant manner. (P<0 05).
