摘要
目的 :比较不同浓度药物对骨髓间质干细胞(BMMSC)体外向心肌细胞诱导分化的影响 .方法 :分离大鼠BMMSC体外培养 ,分别使用终浓度为 1 ,5 ,1 0和 2 0 μmol·L-1 的 5 氮胞苷 (5 aza)对其定向诱导 ,观察细胞形态学变化 ,荧光免疫组织化学方法进行鉴定 .结果 :BMMSC体外经 5 aza诱导 4wk ,肌钙蛋白、肌动蛋白及Connexin4 3表达阳性 ,1 0μmol·L -1 5 aza组及 2 0 μmol·L -1 5 aza组表达较高 ,但 2 0μmol·L -1 5 aza组 5 0 %细胞死亡 .结论 :BMMSC可以在体外经 5 氮胞苷诱导分化为心肌细胞 ,以 1 0 μmol·L -1 5
AIM: To study myogenic differentiation of bone marrow mesenchymal stem cells (BMMSC) in vitro following induction by 5 azacytidine at different concentrations. METHODS: BMMSC from adult rats were cultured in cell culture medium and medium with 5 aza (1, 5, 10 and 20 μmol·L -1 ) for 24 h. After a 4 week culture, the BMMSC were identified by means of immunochemistry technique. RESULTS: BMMSC induced by 5 azacytidine could be identified by the positive staining for Troponin I, Actin and Connexin43. The number of cells stained positively in the 10 μmol· L -1 and 20 μmol·L -1 5 aza was visually greater than that of other groups,but more than 50% of the cells necrosed in 20 μmol·L -1 5 aza. CONCLUSION: BMMSC cultured with 5 aza differentiate into cardiac like muscle cells in culture vitro. The optimal condition for BMMSC to differentiate is 10 μmol·L -1 5 aza.
