摘要
目的 :验证蒙药诃子对草乌煎剂毒动学的影响。方法 :采用动物急性死亡率方法 ,估计乌头碱在小鼠体内的残存量 ,计算其表观半衰期 ,观察了蒙药诃子对草乌煎剂动物体内毒动学的影响。结果 :预先给诃子煎剂(i.g)后再给以生草乌煎剂 (IP)和单独给以生草乌煎剂 (IP)相比 ,草乌在动物体内均呈开放的二室模型。其主要药动学参数变化如下 :k1 2 由 1 .5 97/ h减小到 0 .781 / h;k2 1 由 0 .948/ h增加到 2 .999/ h;k1 0 由 0 .61 2 / h降低为0 .2 96/ h;CL从 0 .61 mg/ h/ (mg/ kg)减少为 0 .2 9mg/ h/ (mg/ kg) ;AUC从 884.7(mg/ kg) h提高到 2 63 2 .4(mg/kg) h。结论 :蒙药诃子对草乌可起到解毒作用 ,它通过和草乌生物碱特异性的结合 ,在体内缓慢释放和消除 ,使毒性缓解。本实验结果为阐明蒙医理论“诃子可解草乌毒”提供实验依据。
Objective: To study the influence of terminalia decoction (TD) on toxicity of aconitum decoction (AD). Methods: Estimation of residual rate and calculation apparent half\|lifetime of aconitum in mouse with the method of acute mortality of animals, to investigate the influence of terminalia on aconitum\'s toxicokinetics. Results: When investigating pharmacokinetical characteristics of TD, an open two\|compartment model was found in mouse with and without TD treatment after being treated with AD. The pharmacokinetical parameters results were as follow: k 12 changed from 1.597/h to 0.781/h, k 21 from 0.948/h to 2.999/h, K 10 from 0.612/h to 0.296/h, CL from 0.61 mg/h/(mg/kg) to 0.29 mg/h/(mg/kg), AUC from 884.7(mg/kg)h to 2632.4(mg/kg)h. Conclusion: The Mongolia medical theory that terminalia can relieve the toxicity of aconitum is reliable.\;
