摘要
目的:探讨哈萨克族食管癌组织中抑癌基因脆性组氨酸三联体(FHIT)基因和O6-甲基鸟嘌呤-DNA甲基转移酶(MGMT)基因启动子区CpG岛异常甲基化程度与食管癌发生的关系。方法:选取30对哈萨克族食管癌组织及癌旁远端正常组织,应用测序法检测食管癌组织与癌旁远端正常组织FHIT和MGMT基因启动子区CpG岛甲基化情况,比较癌组织和正常组织FHIT基因CPG岛9个位点和MGMT基因CPG岛20个位点的甲基化程度。结果:FHIT基因启动子区CpG岛中1、6、8位点(P1=0. 025,P6=0. 013,P8=0. 031)癌组织甲基化程度高于正常组织,MGMT基因启动子区CpG岛中8位点(P8=0. 035)癌组织甲基化程度高于正常组织,差异具有统计学意义(P<0. 05)。结论:哈萨克族食管癌患者癌组织中FHIT基因和MGMT基因的甲基化程度均高于正常组织,提示FHIT基因和MGMT基因启动子区的异常甲基化可能与哈萨克族食管癌的发生有关。
Objective:To study and discuss the relationship between the CpG islands methylation levels of fragile histidine triad(FHIT)gene as well as O6-methylguanine-DNA methyltransferase(MGMT)gene′s promoter regions and the occurrence in Kazak′s esophageal cancer.Methods:Esophageal can-cer tissues and their distal normal tissues from 30 Kazakh patients were selected,and gene sequencing was applied to detect the CpG islands methylation status of FHIT gene and MGMT gene in these tissues.Then the CpG islands methylation levels of FHIT gene at 9 loci and MGMT gene at 20 loci were compared.Results:The CpG island methylation level of FHIT gene in cancer tissues was higher than that in normal tissues at the 1 st,6 th,and 8 th loci(P1=0.025,P6=0.013,P8=0.031),and the CpG island methylation level of MGMT gene in cancer tissues was higher than that in normal tis-sues at the 8 th locus(P8=0.035).The differences are statistically significant(P<0.05).Conclusion:The CpG islands methylation levels of FHIT gene and MGMT gene in cancer tissues were higher than that in normal tissues among Kazakh patients,which suggested that aberrant CpG islands methyl-ation of FHIT gene and MGMT gene′s promoter regions may be correlated with the occurrence of esophageal cancer in Kazakh people.
