摘要
目的探讨环氧化酶-2(COX-2)、上皮钙黏素(E-cad)的mRNA及蛋白在乳腺浸润性导管癌中的表达及临床意义。方法采用原位杂交和免疫组织化学法(SP法)检测30例乳腺纤维腺瘤和70例乳腺浸润性导管癌中COX-2、E-cad的mRNA及蛋白的表达,并进行比较分析。结果 COX-2的mRNA和蛋白在乳腺纤维腺瘤和乳腺浸润性导管癌组织中的阳性率分别为30.0%7、4.2%2,3.3%7、2.8%,二者比较差异均有统计学意义(P<0.01)。E-cad的mRNA和蛋白在乳腺纤维腺瘤和乳腺浸润性导管癌组织中的阳性率分别为93.3%3、5.7%9,0.0%3、2.8%,二者比较差异亦均有统计学意义(P<0.01)。乳腺浸润性导管癌中COX-2的mRNA及蛋白表达均与淋巴结转移有关(P<0.01,P<0.05);E-cad的mRNA及蛋白表达均与淋巴结转移和组织学分期有关(P<0.01,P<0.05)。乳腺浸润性导管癌中COX-2和E-cad的mRNA及蛋白表达的变化趋势均相反,经Spearman相关性检验分析发现两者mRNA及蛋白表达均呈负相关(r=-0.49,P<0.01;r=-0.26,P<0.05)。结论 COX-2的mRNA及蛋白过表达,E-cad的mRNA及蛋白低表达均可促进癌细胞浸润和转移。
Objective To investigate the expressions of COX-2,E-cad gene and protein in invasive ductal carcinoma and to analyze the clinical significance.Methods The expressions of COX-2,E-cad gene and protein in breast fibroadenoma tissues(n=30) and invasive ductal carcinoma tissues(n=70) were detected by in situ hybrid-dization and immunoshistochemisty(SP method).Results The positive rates of COX-2 mRNA and protein in invasive ductal carcinoma were 30.0% and 74.2%,and 23.3% and 72.8% in breast fibroadenoma respectively,and the differences were significant between the two diseases(P<0.01).The positive rates of E-cad mRNA and protein in breast fibroadenoma were 93.3% and 35.7%,but in invasive ductal carcinoma were 90.0% and 32.8% respectively;and the differences were statistically significant between the two diseases(P<0.01).Expressions of COX-2mRNA and protein in invasive ductal carcinoma were related to lymphatic metastasis(P<0.01,P<0.05);The expressions of E-cad mRNA and protein were related to lymphatic metatasis and histopathological grade(P<0.01,P<0.05).The expressions of COX-2 and E-cad mRNA or protein in invasive ductal carcinoma showed negative relationship(r=-0.49,P<0.01;r=-0.26,P<0.05).Conclusion The overexpression of COX-2 mRNA and protein and the low expression of E-cad mRNA and may play important roles in increasing the process of invasion and metastasis of cancer cell.
