新疆哈萨克族ESCC组织LOXL2表达及临床意义 被引量:3

Expression and clinical significance of lysyl oxidase-like 2 in Kazakh esophageal squamous cell carcinoma

摘要 目的哈萨克族食管癌属于新疆地区特高发疾病,其发病率及死亡率均高于全国平均水平。近来越来越多的研究表明,赖氨酰氧化酶样蛋白2(lysyl oxidase-like 2,LOXL2)与肿瘤的侵袭、转移及不良预后有密切关系。本研究探讨LOXL2在新疆哈萨克族食管鳞癌(esophageal squamous cell carcinoma,ESCC)患者中的表达与临床病理特征相关性及与预后的关系。方法检索Oncomine数据库中LOXL2mRNA在ESCC患者中的表达情况,收集2009-03-01-2012-03-31新疆医科大学附属肿瘤医院胸外科80例新疆哈萨克族ESCC患者手术切除标本,免疫组织化学检测LOXL2在80例新疆哈萨克族ESCC患者中蛋白表达,并分析其表达与临床病理特征及总生存时间(overall survival,OS)的关系。结果 Oncomine数据库中共有2个研究涉及ECT2在ESCC组织和正常组织中mRNA表达,2组微阵列数据集中,LOXL2mRNA在ESCC组织中的转录水平均显著高于正常组织,P=3.95×10^(-16),P=6.87×10^(-7)。80例ESCC组织标本及癌旁组织LOXL2蛋白阳性表达率分别为55.0%和28.7%(χ2=4.665,P=0.031),且LOXL2蛋白表达与ESCC的肿瘤分化程度(χ~2=9.126、P=0.010)、淋巴结转移(χ~2=4.393、P=0.036)及TNM分期(χ~2=6.519、P=0.011)呈正相关,与患者性别、年龄、浸润深度及CEA水平无明显相关性,P>0.05。LOXL2阳性表达阴性表达的患者5年OS分别为6.7%和35.8%,差异有统计学意义,χ~2=4.612,P=0.032。结论 LOXL2蛋白在新疆哈萨克族ESCC组织中表达明显增高,其蛋白阳性表达与肿瘤分化程度、淋巴结转移及TNM分期密切相关,LOXL2在新疆哈萨克族ESCC的发生发展中起重要作用,可以作为反映新疆哈萨克族食管癌生物学行为及预后的有效指标。 OBJECTIVE Kazakh esophageal cancer is a high incidence of disease in Xinjiang,and the morbidity and mortali- ty are higher than the national average. More and more studies have shown that lysyl oxidase-like 2 (I.OXL2) is closely related to tumor invasion, metastasis and poor prognosis. The aim of this study was to investigate the expression of lysyl oxidase-like 2 (LOXL2) in Kazakh esophageal squamous cell carcinoma (ESCC) and its correlation to clinicopathologic features and prognosis. METHODS Data about the expression of LOXL2 mRNA in ESCC were retrieved from the ONCOMINE database. Totally 80 ca ses of Xinjiang Kazakhstan esophageal squamous cell carcinoma surgical resection specimens from department of thoracic surgery, affiliated tumor hospital, Xinjiang Medical University during March 1, 2009 March 31, 2012 were collected. Expression of LOXL2 was detected in 80 samples of Kazakh ESCC by immunohistochemical method. The associations between LOXL2 expres- sion and clinicopathologic features, overall survival were evaluated. RESULTS The expression of LOXL2 mRNA in ESCC was significantly higher than that in normal tissues in the Oncomine database(P = 3.95 X 10 16, p = 6.87 X 10 -7 ), Expression of LOXL2 protein was significantly higher in Kazakh ESCC than in matched tumor adjacent tissues (55.0 % vs 28.7 %, Xz = 4. 665, P=0. 031). Up-regulation of LOXL2 were related with tumor differentiation (X2 = 9. 126, P= 0. 010), lymph node metastasis(X2 =4. 393,P〈0. 036) and TNM stage (2 =6. 519,P=0.01D in Kazakh ESCC, but they were not correlated with the gen- der, age, invasion of depth and CEA level(P〈0.05). The 5-year overall survival rate in patient with positive LOXL2 expression was 6.7%. However, 5-year overall survival rate in patient with negative LOXL2 expression was 35. 8% (X2= 4. 612.P=0. 032). CONCLUSIONS The expression of LOXL2 protein is up-regulated in Kazakh ESCC. Up-regulation of LOXL2 was re- lated with tumor differentiation, lymph node metastasis TNM stage in Kaz
