大鼠脑损伤后脑皮质及血清5—HT和5—HIAA含量及赛庚啶对其的影响 被引量:3

Effect of cyproheptadinum on changes of 5-HT and 5-HIAA in brain cortex and serum following brain injury

摘要 通过大鼠脑落体损伤模型,采用荧光分光光度仪检测脑损伤后不同时间脑皮质和血清中5-羟色胺(5-HT)及其代谢产物5-羟吲哚醋酸(5-HIAA)含量。观察了用盐酸赛庚啶治疗后对5-HT和5-HIAA含量的影响。结果表明,脑皮质中5-HT和5-HIAA含量均升高。伤后48h,脑皮质中5-HT升高达高峰,为4.29±0.44μg,是对照值的4.8倍;5-HTAA于伤后24h达高峰,为5.48±0.41μg/g,是对照值的5.8倍。血清中5-HT变化不明显,5-HIAA亦明显升高。应用5-HT受体阻断剂赛庚啶治疗后,脑皮质中5-HT下降为1.32±0.09μg/g,5-HIAA含量下降为1.36±0.10μg/g;血清中5-HIAA含量也较明显降低为0.27±0.03μg/ml。 The changes of 5-hydroxytryptamine(5-HT ) and 5-hydroxyindoleacetic acid ( 5-HIAA) levels in brain cortex and serum following brain injury were stu-died with spectrofluorometer.Cyproheptadinum,a receptor antagonist of 5-HT,was employed to show its effect on changes of 5-HT and 5-HIAA in brain cortex and serum in the present study. The results showed that the levels of 5-HT and 5HIAA in the brain cortex increased. The augmentation were most remarkable at 24 and 48 hours after injury.The values were 4. 29±0.44μg/g and 5.48±0.41μg/g respectively, which were 4.8 and 5.8 times of the control values. The changes of 5-HT in serum was not evident,but the levels of 5-HIAA in serum was also higher than the control value.Cyproheptadinum showed the effect to reduce the levels of 5-HT and 5-HIAA in this study. The values of 5-HT in brain cortex was 1.32± 0.09μ/g, and the value of 5-HIAA in brain cortex was 1.36±0.10μ/g after the administration of cyproheptadinum. The level of 5-HIAA in serum was decreased significantly, too. It was 0. 27±0.03μg/ml.
