摘要
本研究以自有亲本材料B0与地方品种泉烟、柳烟杂交后与云85、红花大金元组配的三交材料为试材,用辐照剂量为100 Gy的电子束对其进行辐照处理,对辐照后代烟草材料小孢子培养技术进行研究。结果表明:不同基因型材料对游离小孢子培养的敏感性不同,所有的材料中产胚率最高的是(B0×柳烟)×云85组合,产胚率17.2;在材料4和材料6中随着秋水仙碱浓度的提高,两个材料的加倍率均增加,在浓度为100 mg/L时两个材料的加倍率最大。
The parental material B0 specific to Tobacco Technology Center of Sichuan Province is used to cross with the local varieties like Quanyan, Liuyan. And then it is combined with Yun85 and Honghua Dajinyuan,which is used to be the experimental material. By using electronic beam(100 Gy) to irradiate the experimental material, the offspring of the irradiated tobacco is analyzed by the technology of isolated microspore culture. The results indicate that different genetic materials have different sensibility to the isolated microspore culture.Among all of the materials, the combination of(B0×Liuyan)×Yun85 shows the highest embryogenic rates, which is up to 17.2. In material 4 and material 6, with the increase of concentration of colchicines, the doubling rate of both materials goes up. When the concentration comes to 100 mg/L, the doubling rate will be the highest.
