摘要
目的通过体外人脐静脉内皮细胞和海绵植入小鼠模型对丹参酮Ⅱ_A磺酸钠的促进血管新生作用进行研究。方法体外采用人脐静脉内皮细胞HUVEC-12诱导模型,利用细胞迁移及小管生成实验评价丹参酮Ⅱ_A磺酸钠促血管新生的作用影响;体内实验采用海绵植入小鼠模型,通过QPCR测定不同剂量丹参酮Ⅱ_A磺酸钠对VEGF-A mRNA表达的影响,采用免疫荧光染色法观察不同剂量丹参酮Ⅱ_A磺酸钠对微血管生成及VEGF蛋白释放的影响。结果丹参酮ⅡA磺酸钠40、80μg/m L剂量组的迁移率显著高于对照组(P<0.05、0.01);丹参酮Ⅱ_A磺酸钠与形成的结节数目呈现一定剂量相关性,且丹参酮Ⅱ_A磺酸钠80μg/mL剂量组的小管生成节结数显著高于对照组(P<0.05)。丹参酮ⅡA磺酸钠12、24mg剂量组VEGF-Am RNA相对表达值显著高于对照组(P<0.01),而丹参酮Ⅱ_A磺酸钠12mg剂量组作用最强,优于阳性药丹参多酚酸盐。丹参酮ⅡA磺酸钠12、16、24mg剂量组的VEGF蛋白表达量明显优于对照组(P<0.05、0.01),丹参酮Ⅱ_A磺酸钠12mg剂量组,VEGF蛋白表达量最高,荧光面积最大,优于阳性药丹参多酚酸盐。结论丹参酮Ⅱ_A磺酸钠具有促血管新生作用,其作用机制可能通过上调VEGF-A mRNA的表达来影响。
Objective To study the effect of angiogenesis effect of sodium tanshinoneⅡ_A sulfonate on human umbilical vein endothelial cell in vitro and sponge implantation in rats.Methods HUVEC-12 induction model was selected in vitro,and the effect of sodium tanshinoneⅡ_A sulfonate on angiogenesis was evaluated by cell migration and tubule formation.Sponge implantation assay was selected in vivo,and the effect of sodium tanshinoneⅡ_A sulfonate in different doses on the expression of VEGF-A mRNA was detected by QPCR.The effects of sodium tanshinoneⅡ_A sulfonate in different doses on microvascular production and VEGF protein release were observed by immunofluorescence staining.Results The migration rate in sodium tanshinoneⅡ_A sulfonate 40 and 80μg/mL group were significantly higher that in the control group(P<0.05,0.01).Sodium tanshinoneⅡ_A sulfonate had the dosage association with the number of tubule formation,and the number of tubule formation in sodium tanshinoneⅡ_A sulfonate 80μg/mL group was significantly higher that in the control group(P<0.05).VEGF-A mRNA expression in sodium tanshinoneⅡ_A sulfonate 12 and 24 mg group were significantly higher that in the control group(P<0.01).VEGF-A mRNA expression in sodium tanshinoneⅡ_A sulfonate12 mg group was the highest,and higher than that in salvianolate group.VEGF protein expression in sodium tanshinoneⅡ_A sulfonate12,16,and 24 mg group was significantly higher that in the control group(P<0.05,0.01).VEGF protein expression in sodium tanshinoneⅡ_A sulfonate 12 mg group was the highest,and fluorescence area was the biggest,and better than that in salvianolate group.Conclusion Sodium tanshinoneⅡ_A sulfonate has the effect of promoting angiogenesis,which is related to promoting the expression of VEGF-A mRNA mainly.
