摘要
目的观察岷当归对高脂饲料(HDF)联合链脲佐菌素(STZ)诱导的Ⅱ型糖尿病小鼠血清白细胞介素-6(IL-6)、白细胞介素-22(IL-22)和肿瘤坏死因子-α(TNF-α)含量的影响。方法将40只昆明种小鼠随机分为空白组、模型组、防治组和治疗组,每组10只。防治组提前2周灌胃当归水煎液,2周后除空白组外,其他组采用HDF联合STZ腹腔注射造模,造模成功后防治组和治疗组灌胃当归水煎液,其他2组继续灌胃生理盐水,连续灌胃4周。在处死小鼠前1天,各组小鼠进行葡萄糖耐量(OGTT)试验,经药物干预后采用酶联免疫法检测各组小鼠血清IL-6,IL-22和TNF-α的含量。结果与模型组比较,防治组和治疗组小鼠OGTT值,血清IL-6,IL-22和TNF-α含量均降低,差异均有统计意义(P<0.05);且防治组小鼠OGTT值,血清IL-6,IL-22和TNF-α含量下降更明显,差异均有统计意义(P<0.05)。结论岷当归可以调节HDF联合STZ诱导的Ⅱ型糖尿病小鼠血清IL-6,IL-22和TNF-α的分泌,缓解Ⅱ型糖尿病小鼠的炎症反应,从而起到调节Ⅱ型糖尿病小鼠免疫平衡的作用。
Objective To observe the effect of angelica in Minxian County on the contents of IL-6,IL-22 and TNF-α in serum of mice with type Ⅱ diabetes induced by high-fat diet and streptozotocin. Methods 40 mice were randomized into blank group,model group,prevention group and treatment group,10 for each. The prevention group was given angelica decoction intragastrically 2 weeks ahead. Except blank control group,other groups were modeled through high-fat diet and streptozotocin. Model mice in prevention group and treatment group were given gastric perfusion of angelica decoction and other two groups were given normal saline,lasting for 4 weeks. OGTT was run in all mice day-ahead before they were killed. The contents of IL-6,IL-22 and TNF-α were detected with ELISA after drug intervention. Results Compared with model group,all contents in prevention group and treatment group declined evidently with statistical significance( P < 0. 05) and that in prevention group dropped more,there was a difference( P < 0. 05). Conclusion Angelica in Minxian County can regulate the secretion of IL-6,IL-22 and TNF-α in serum as well as remit the inflammatory reaction. The drug plays a role in adjusting the immune balance in mice with typeⅡdiabetes.
