摘要
目的探讨关节腔内注射不同浓度木瓜蛋白酶建立小鼠膝骨关节炎模型的可行性。方法健康10周龄雄性昆明小鼠80只,体重(45±3)g,随机分为2%木瓜蛋白酶组、4%木瓜蛋白酶组、8%木瓜蛋白酶组和对照组。实验第1、3、5天分组行右膝关节腔内注射2%、4%、8%(w/v)木瓜蛋白酶和0.3mol/LL-半胱氨酸的混合溶液0.25 m L/kg或等量生理盐水。记录小鼠一般情况及右膝关节宽度。所有小鼠首次注射1周后处死,取关节行常规HE染色及甲苯胺蓝染色,镜下观察并评分。结果造模组右膝关节出现肿胀、功能障碍;右膝关节软骨染色变浅、软骨层变薄、基质分解破坏等病理学改变,其严重程度与木瓜蛋白酶浓度呈正相关;对照组无上述改变。结论膝关节腔内注射2%、4%、8%(w/v)木瓜蛋白酶和0.3mol/L L-半胱氨酸的混合溶液0.25 m L/kg可建立小鼠不同严重程度的膝骨关节炎,周期短、成功率高。
Objective To determine the feasibility of the knee osteoarthritis( OA) models in mice with different concentrations of papain mixed solution. Method Eighty Kunming mice,weight( 45 ± 3) g,were randomly divided into four groups of 20 each( 2% papain group,4% papain group,8% papain group,and control group) and given injections into the right knees on the 1st,3rd,5th day respectively. The mixed solution,composed of 2%,4%,8%( w / v) papain and 0. 3mol / L l-cysteine,and 0. 9% Nacl were applied to experimental groups and control group respectively at the dose of 0. 25 ml / kg. All mice were sacrificed at one week after the initiation of papain injection and evaluated through recording the width of knee joints,performing the morphological observation( toluidine bluc staining & HE staining) and histological evaluation of articular cartilage. Results Swelling and activities diminishment could be observed in the knee joints of experimental groups. The degenerative changes were demonstrated in the knee joints of all experimental groups,such as shallow staining,thinner articular cartilage and destroyed cartilage matrix,and inflammation. All these changes were much worse with increased concentration.Conclusion Different severities of OA can be established through intra-articular injections of 2%,4%,8% papain and 0. 03 mol / L lcysteine at the dose of 0. 25 ml / kg. These models are of the characters of short period and a good reproducibility.
