摘要
目的研究雷帕霉素对鼻咽癌肿瘤干细胞CNE2增殖的影响。方法将鼻咽癌肿瘤干细胞CNE2细胞随机分为对照组(0.9%NaCl)和低、中、高剂量实验组(0,50,100,200 ng·mL-1雷帕霉素)。给药48 h后,以四甲基偶氮唑蓝(MTT)法检测各组细胞增殖抑制情况;用Western blot检测各组CNE2细胞哺乳动物雷帕霉素靶点通路(mTOR)及磷酸化mTOR(p-mTOR)蛋白表达情况。结果低、中、高剂量实验组的细胞增殖抑制率分别为(24.62±7.18)%,(38.46±7.49)%,(49.23±7.91)%,差异均有统计学意义(均P <0.05);对照组及低、中、高剂量实验组CNE2细胞mROT蛋白相对表达量分别为1.02±0.33,0.87±0.25,0.03±0.01,0.02±0.01;p-mROT蛋白相对表达量分别为0.94±0.29,0.57±0.21,0.06±0.02,0.04±0.01。低、中、高剂量实验组的上述指标分别与对照组比较,差异均有统计学意义(均P <0.05)。结论雷帕霉素可通过下调mTOR及p-mTOR表达,降低mTOR信号通路活性,进而显著抑制鼻咽癌肿瘤干细胞CNE2增殖,达到治疗鼻咽癌的目的。
Objective To explore the effect of rapamycin on the proliferation of nasopharyngeal carcinoma cell line CNE-2.Methods The CNE2 cells were randomly assigned to control group(0.9%NaCl)and low-/middle-/high-dose experimental group(50,100,200 ng·mL-1 of rapamycin)and treated for 48 h.The proliferation inhibition of each group were detected by Methylthiazolyldiphenyl-tetrazolium(MTT)method.The expression of mammalian target of rapamycin(mTOR)and phosphorylated mTOR(p-mTOR)protein were detected by Western Blot.Results There were statistically significant differences among the low-/middle-/high-dose experimental group in proliferation inhibition rates[(24.62±7.18)%vs(38.46±7.49)%vs(49.23±7.91)%]at 48 h after treatment(P<0.05).The relative expression of mROT protein of CNE2 cells in control group and low-/middle-/high-dose experimental group were 1.02±0.33,0.87±0.25,0.03±0.01,0.02±0.01,respectively;the relative expression of p-mROT of CNE2 cells were 0.94±0.29,0.57±0.21,0.06±0.02,0.04±0.01 respectively.There were statistically significant differences between the control group and the low-/middle-/high-dose experimental group in the relative expression of mTOR and p-mTOR protein of CNE2 cells(P<0.05).Conclusion Rapamycin can inhibit the proliferation of nasopharyngeal cancer stem cells CNE2 through the down-regulation of mTOR and p-mTOR expression,reduce the activity of mTOR signal path,achieve the goal of the treatment of nasopharyngeal carcinoma.
