摘要
目的探究桔梗皂苷-D对胃癌BGC823细胞增殖、侵袭及迁移能力的影响。方法将胃癌BGC823细胞随机分为对照组和低、中、高3个剂量实验组,对照组予以0.9%Na Cl,低、中、高剂量实验组分别以终浓度为5,10,20μmol·L-1桔梗皂苷-D处理。给药48 h后,噻唑蓝(MTT)法检测BGC823细胞增殖情况,Transwell小室实验及划痕法分别检测BGC823细胞侵袭及迁移能力。结果给药48 h后,低、中、高剂量实验组BGC823细胞增殖抑制率分别为(17.09±0.92)%,(32.03±0.87)%,(45.04±0.88)%;给药48 h后,对照组、低、中、高剂量实验组侵袭细胞数量分别为(348.74±24.79),(120.33±16.54),(82.05±10.21),(49.23±9.64)个,划痕愈合率分别为(67.01±3.72)%,(36.23±3.54)%,(27.14±3.20)%,(19.11±3.01)%。差异均有统计学意义(均P <0.05)。结论桔梗皂苷-D可有效抑制胃癌BGC823细胞增殖,降低其侵袭及迁移能力,且具有显著的剂量依赖性。
Objective To explore the effect of platycodin D on the proliferation,invasion and migration capacity of gastric cancer BGC823 cell.Methods Gastric cancer BGC823 cells were randomly divided into control group and low-/middle-/high-dose experimental group.The control group was given 0.9%NaCl,while low-/middle-/high-dose experimental group were treated with platycodon saponin-D at the final concentration of 5,10 and 20μmol·L-1,respectively,and each group was treated for 24,48 h respectively.The proliferation inhibition of BGC823 cells were detected by methyl thiazolyl tetrazolium(MTT),the invasion and migration capacity of BGC823 cells were detected by transwell chamber test and Wound Healing method respectively.Results The proliferation inhibition rate of BGC823 cell in the low-/middle-/high-dose experimental group were(17.09±0.92)%,(32.03±0.87)%,(45.04±0.88)%at 48 h after intervention.The number of invasive cells in the control group,low-/middle-/high-dose experimental group at 48 h after intervention were 348.74±24.79,120.33±16.54,82.05±10.21,49.23±9.64,respectively,the healing rate of scratches was(67.01±3.72),(36.23±3.54),(27.14±3.20),(19.11±3.01)%.The difference was statistically significant(all P<0.05).Conclusion Platycodin D can inhibit the proliferation of gastric cancer BGC823 cells effectively,and reduce the invasion and migration capacity,which has significant dose dependence.
