基因枪介导bar基因表达盒转化甜玉米初步研究

摘要 以超甜玉米优良自交系1132幼胚的愈伤组织为受体材料,采用基因枪介导法将bar基因表达盒(启动子+基因+终止子)导入胚性愈伤组织,经选择培养、胚状体诱导、再生等过程,获得38个株系共101株转基因玉米。对转基因植株的叶片涂抹除草剂basta进行抗性鉴定,其中有8个株系共22株表现抗性;进一步进行PCR分析,结果表明,其中有6个株系共15株为阳性。试验结果初步证明bar基因已整合到1132的基因组中,转化效率为3%。转基因表达盒完整性分析表明,bar基因表达盒在受体基因组中的结构是完整的。 Herbicide reisistance bar gene expression cassette(containing the promoter,coding region and terminator) was introduced into immature embryo callus of the sweet corn inbred line 1132 through microprojectile bombardment.After selection on media supplemented with different concentration PPT,38 independent transgenic lines were obtained.The PPT test of leaf tissue indicated that 22 transgenic plant lines were resistant to PPT.PCR analysis showed that 6 independent transgenic lines were positive and the structur of bar gene cassette was integrated in the receptor genome.The transformation efficiency was 3% in this research.
