摘要
目的 :寻找提高肝癌诊断率的标记物。方法 :应用重组克隆抗原表达的血清学鉴定 (SEREX)技术筛查获取新的肝癌抗原 (HCC- 1- 8和 HCC- 3- 13) ,对 2 3例健康人及 12例肝癌患者进行上述两种抗原相应抗体的免疫检测及甲胎蛋白 (AFP)定性检测 ,并比较检测结果。结果 :2 3例健康人抗 HCC- 1- 8及抗 HCC- 3- 13为阴性 ,AFP定性检测为阴性 ;12例肝癌患者抗 HCC- 1- 8及抗 HCC- 3- 13阳性检出数分别为 10例和 11例 ,AFP阳性者 6例 ,阴性者 6例 ;其中 6例 AFP阳性者抗 HCC-1- 8阳性反应 5例 ,抗 HCC- 3- 13阳性反应 6例 ;6例 AFP阴性者抗 HCC- 1- 8阳性反应 5例 ,抗 HCC- 3- 13阳性反应者 5例。结论 :HCC- 1- 8和 HCC- 3- 13相应抗体在 AFP阴性健康人未检出而在肝癌患者阳性检出数高于 AFP。初步提示肝癌病人血清中两种新肝癌抗原的相应抗体检测较 AFP灵敏 。
Objective:To find serum markers that can improve diagnostic rate of hepatocellular carcinoma (HCC) Methods:Two new HCC antigens (HCC 1 8 and HCC 3 13) were obtained from HCC cDNA library with serological identification of antigens by recombinant expression cloning (SEREX) technique; they reacted to the sera of 23 health and 12 HCC cases who had been tested α fetus protein(AFP) Results:Antibody negative reaction to HCC 1 8 and HCC 3 13 were in all of 23 health whose AFP showed negative (-) The cases of the antibody positive response to HCC 1 8 were 10 in 12 HCC among there were 5 cases in 6 HCC whose AFP showed positive (+) and 5 cases in 6 HCC whose AFP showed (-); that to HCC 3 13 were 11 in 12 HCC, there were 5 cases in 6 AFP (+) and all cases in 6 AFP (-) Conclusion:Positive response rate of antibodies to HCC 1 8 and HCC 3 13 was higher than that of AFP in HCC It is considered that antibodies to HCC 1 8 and HCC 3 13 act as the serum markers that can raise the rate in diagnosing of HCC
