血人参原花青素合成关键酶基因IsANR、IsLAR的鉴定及分析

Identification and Analysis of IsANR and IsLAR as Key Enzyme Gene in Biosynthesis of Procyanidins in Indigofera stachyoides

摘要 花青素还原酶(ANR)及无色花青素还原酶(LAR)是原花青素单体生物合成过程中两个关键限速酶,对原花青素的生物合成及积累具有重要作用。本研究以黔产血人参为材料,根据其转录组测序结果,采用生物信息学方法从数据库中鉴定IsANR和IsLAR基因,并分析其蛋白结构及表达模式,揭示其与血人参原花青素积累的关系。本研究从血人参中鉴定出了2个IsANRs、2个IsLARs基因,其中IsLAR1蛋白的氮端具有叶绿体转运体。IsANR2基因的编码区中存在终止子,导致蛋白翻译提前终止。二级结构分析揭示IsANR1比IsANR2有较高比例的α-螺旋,较低的无规卷曲,IsLAR1比IsLAR2有较高的β-转角。IsANR2主要在叶中表达,IsANR1、IsLAR1在血人参根部具有较高的表达量,且IsLAR1和IsLAR2在根中的表达量随年限的增加而上升。因此,血人参根部原花青素的积累可能与LAR基因(尤其是IsLAR1)的上调表达有密切的关系。本研究结果不仅有助于揭示根部原花青素生物的合成机制,同时也为血人参药材的质量控制提供了理论参考。 Anthocyanin reductase(ANR)and leucoanthocyanidin reductase(LAR),two key rate limiting enzymes in the biosynthesis of proanthocyanidins,play an important role in the biosynthesis and accumulation of proanthocyanidins.IsANRs and IsLARs were identified from the transcriptome of Indigofera stachyoides by bioinformatics method,and structure and expression pattern of them were analyzed to reveal their function during the accumulation of proanthocyanidins in I.stachyoides.In this study,2 IsANRs and 2 IsLARs were identified respectively and a chloroplast transporter peptide was discovered from N-terminal of IsLAR1.Terminators exist in the coding region of IsANR2 gene,which leads to premature termination of protein translation.Secondary structure analysis revealed that IsANR1 had a higher rate ofα-helix and a lower random coil than IsANR2,and IsLAR1 had a higherβ-turn than IsLAR2.IsANR2 was mainly expressed in leaves.High transcripts of IsLAR1 and IsLAR2 were detected in root of I.stachyoides and increased along with the age.Therefore,the accumulation of proanthocyanidins in I.stachyoides roots is possibly related to the up-regulated expression of LAR genes(especially IsLAR1).The results may not only help to explore the biosynthetic mechanism of proanthocyanidins in roots,but also provide theoretical reference for the quality control of I.stachyoides.
