摘要
建立超高效液相色谱法测定固态食品中泛酸含量的分析方法。样品中泛酸经无水甲醇提取、氮气吹干、纯水复溶,过滤除杂后,以磷酸二氢钾溶液和乙腈为流动相,通过Waters,CORTECS UPLC T3色谱柱梯度洗脱,流速0.35 ml/min,进样量2μL,柱温30℃,检测波长200 nm,使用超高效液相色谱测定分析。结果显示,泛酸在0.001~0.031μg/L浓度范围内具有良好的线性关系,相关系数为1.0,方法检出限为0.025 mg/100 g,定量限为0.08 mg/100 g,泛酸在婴幼儿配方食品、婴幼儿辅助食品、代餐营养食品等固态样品中测定结果的相对标准偏差值为0.9%~4.1%;加标回收率均大于90%(n=6)。针对含有蛋白、肽,淀粉等复杂基质的固态配方食品,本液相色谱法的检测一致性较好,分离度、精密度、准确性等均符合要求。有效解决复杂基质、杂质干扰下检测结果不稳定、平行差的问题。
To establish an analytical method for determining the content of pantothenic acid in solid food by ultra-high performance liquid chromatography.Pantothenic acid in the sample was extracted with anhydrous methanol,dried with nitrogen,and dissolved in pure water.Afterfiltering out impurities,potassium dihydrogen phosphate solution and acetonitrile were used as mobile phases.The mixture was eluted using a Waters,CORTECS UPLC T3 column gradient with aflow rate of 0.35ml/min,injection volume of 2μL,column temperature of 30℃,detection wavelength of 200nm,and analyzed using ultra-high performance liquid chromatography.Pantothenic acid has a good linear relationship within the concentration range of 0.001~0.031μg/L,with a correlation coefficient of 1.0.The limits of detection is 0.025 mg/100 g and the limits of quantification is 0.08 mg/100 g.The relative standard deviation of pantothenic acid in solid samples such as infant formula food,infant supplement food,and meal replacement nutrition food is 0.9%~4.1%;The recovery rates of spiked samples were all greater than 90%(n=6).For solid formula foods containing complex matrices such as proteins,peptides,and starch,the detection consistency of this liquid chromatography method is good,and the separation,precision,and accuracy all meet the requirements.Effectively solve the problems of unstable detection results and parallelism under complex matrix and impurity interference.
