番石榴叶总黄酮对MDA-MB231凋亡相关因子Caspase-3及Survivin的影响

摘要 目的 研究番石榴叶总黄酮对MDA-MB231凋亡相关因子Caspase-3及Survivin的影响.方法 以番石榴叶总黄酮为原料,采用体外细胞培养方法,MTT法检测其对MDA-MB231肿瘤细胞株的生长抑制率,流式细胞仪检测总黄酮作用下MDA-MB231细胞的凋亡率,ELISA方法检测Caspase-3及Survivin蛋白表达情况.结果 番石榴叶总黄酮对乳腺癌MDA-MB231细胞的增殖具有显著抑制作用,随着番石榴叶总黄酮给药剂量的增加,对MDA-MB231肿瘤细胞株的凋亡相关因子Caspase-3具有激活作用,对Survivin因子具有下调作用,分别与对照组比较,差异有统计学意义(P<0.05),凋亡率随着总黄酮给药浓增加而增加(药物浓度:低剂量80μg/ml,中剂量160μg/ml,高剂量320μg/ml),MDA-MB231细胞的凋亡率逐渐增加(凋亡率:16.0%,23.5%,26.3%).结论 番石榴叶总黄酮对MDA-MB231细胞的增殖具有显著抑制作用,对凋亡相关因子Caspase-3具有激活作用,对Survivin因子具有下调作用. Objective To study the effect of Guava leaves flavonoids on the proliferation inhibition of MDA-MB231 breast cancer cells and the expression levels of survivin and Caspase-3.Methods The MTT method was used to detect the inhibition rate of flavonoids to the MDA-MB231 cells,the sandwich enzyme-linked immuno-absorbent assayed(ELISA)was used to detect the apoptosis-rate and the level of survivin and Caspase-3 in MDA-MB231 cells influenced by flavonoids.Results The guava leaves flavonoids has a significant inhibitory effect on the proliferation of MDA-MB231 cells of breast cancer.With the increase of the dosage of total flavone of guava leaf,it has an activation effect on caspase-3,an apoptosis related factor of MDA-MB231 tumor cell line,and a down-regulation effect on survivin factor.There is a statistically significant difference between the two groups(P<0.05).The apoptosis rate increases with the increase of the concentration of total flavone(drug concentration:80μg/ml at low dose,160μg/ml at medium dose,320μg/ml at high dose).The apoptosis rate of MDA-MB231 cells gradually increases(apoptosis rate:16.0%,23.5%,26.3%).Conclusion The flavonoids can inhibit the MDA-MB231 cancer cells significantly and the inhibition rate were increased with the treatment concentration,also can increase the levels of Caspase-3 and lower the level of survivin.
