摘要
目的 探讨结直肠癌(CRC)组织赖氨酸特异性脱甲基酶3A(KDM3A)表达与临床病理特征及预后的关系。方法 选择2018年1月至2020年12月河南中医药大学第三附属医院收治的86例行手术治疗的CRC患者为研究对象。应用免疫组织化学的方法检验KDM3A在CRC组织和癌旁组织中的表达水平,比较不同临床病理特征CRC患者KDM3A蛋白表达的差异;随访3年,分析KDM3A蛋白表达与CRC患者预后的关系。结果 免疫组化结果显示,CRC组织中KDM3A高表达率高于癌旁组织,差异有统计学意义(P<0.05);TNMⅢ期、低分化、有神经侵犯及淋巴结转移阳性CRC患者癌组织内KDM3A高表达率高于TNMⅠ、Ⅱ期、高中分化、无神经侵犯及淋巴结转移阴性患者,差异均有统计学意义(P均<0.05);86例CRC患者中随访过程中死亡29例,存活57例。KDM3A高表达组与低表达组生存时间、3年累积生存率比较,差异有统计学意义(P<0.05)。结论 CRC组织中KDM3A呈高表达,且与CRC的临床病理特征和预后紧密相关,可作为有价值的生物标志物用于CRC的诊断和预后评估。
Objective To investigate the relationship between lysine-specific demethylase 3A(KDM3A)expression and clinicopathological features and prognosis in colorectal cancer(CRC)tissue.Methods A total of 86 CRC patients who underwent surgical treatment in the Third Affiliated Hospital of Henan University of Chinese Medicine from January 2018 to December 2020 were selected as the study objects.Immunohistochemistry was used to detect the expression of KDM3A in CRC tissues and adjacent tissues,and the difference of KDM3A protein expression in CRC patients with different clinicopathological characteristics was compared.The relationship between the expression of KDM3A protein and the prognosis of CRC patients was analyzed after 3 years of follow-up.Results Immunohistochemical results showed that the high expression rate of KDM3A in CRC tissues was higher than those in adjacent tissues,and the differences were significant(P<0.05).The high expression rate of KDM3A in cancer tissues of CRC patients with TNM stage II,low differentiation,nerve invasion and positive lymph node metastasis were higher than those of CRC patients with TNM stage I,II,high differentiation,no nerve invasion and negative lymph node metastasis,with significant differences(P<0.05).Among the 86 CRC patients,29 patients died and 57 patients survived during follow-up(P<0.05).There was significant difference in the survival time and 3-year culmulative survival rate between the KDM3A high expression group and KDM3A low expression group(P<0.05).Conclusions KDM3A is highly expressed in CRC tissues,and is closely related to the clinicopathological features and prognosis of CRC,which can be used as a valuable biomarker for the diagnosis and prognosis evaluation of CRC.
