摘要
目的:建立基于标准汤剂的青钱柳配方颗粒的质量标准。方法:收集15批不同产地的青钱柳药材,制备15批青钱柳标准汤剂及3批配方颗粒,采用高效液相色谱法(HPLC)测定青钱柳标准汤剂和配方颗粒中绿原酸的含量,并建立两者特征图谱;采用薄层色谱法(TCL)对两者进行定性鉴别。结果:15批青钱柳标准汤剂绿原酸含量为6.51~17.67 mg/g,转移率范围31.1%~88.4%;3批颗粒含量分别为6.91、7.13、6.99 mg/g,转移率分别为41.7%、57.4%、51.7%。建立了青钱柳标准汤剂和配方颗粒特征图谱,共标定了6个共有峰,指认了3个成分,其中峰1为新绿原酸,峰2(S)为绿原酸,峰3为隐绿原酸。TLC法中青钱柳标准汤剂和配方颗粒与对照品色谱相应的位置上,显相同颜色的荧光斑点。结论:本研究建立的基于标准汤剂的青钱柳配方颗粒质量标准合理可行,可为青钱柳配方颗粒的质量控制及工艺研究提供参考。
Objective:To establish quality evaluation method of Qingqianliu(Cyclocarya paliurus)dispensing granules based on standard decoction.Methods:15 batches of Qingqianliu(Cyclocarya paliurus)were collected from different habitats,15 batches of Qingqianliu(Cyclocarya paliurus)standard decoction and three batches of dispensing granules were prepared.The content of chlorogenic acid in Qingqianliu(Cyclocarya paliurus)standard decoction and dispensing granules was determined by HPLC,and their characteristic maps were established.TCL method was used to identify them qualitatively.Results:The chlorogenic acid content of 15 batches of Qingqianliu(Cyclocarya paliurus)standard decoction was 6.51-17.67 mg/g,and the transfer rate ranged from 31.1%to 88.4%.The particle contents of three batches are 6.91,7.13 and 6.99 mg/g;the transfer rates are 41.7%,57.4%and 51.7%.The characteristic maps of Qingqianliu(Cyclocarya paliurus)standard decoction and dispensing granules were established,and six common peaks were calibrated and three components were identified,among which peak 1 was Neochlorogenic acid,Peak 2(S)was Chlorogenic acid and Peak 3 was Cryptochlorogenic acid.In TLC method,the standard decoction and dispensing granules of Qingqianliu(Cyclocarya paliurus)show fluorescent spots of the same color at the corresponding positions of the chromatogram of the reference substance.Conclusion:The quality standard of Qingqianliu(Cyclocarya paliurus)dispensing granules based on standard decoction established in this study is reasonable and feasible,which can provide reference for the quality control and process research of Qingqianliu(Cyclocarya paliurus)dispensing granules.
