基于体外生物活性的中国地表水中G蛋白偶联受体药物筛选

In vitro biological activity based prioritization of G protein-coupled receptor-acting pharmaceuticals in surface water in China

摘要 低浓度(ng/L至μg/L)的药物在环境水体中被广泛检出,其对水生生物存在潜在的生态风险,进而受到人们的关注.G蛋白偶联受体(GPCR)是一类在人体与动物体内高度保守的跨膜蛋白,约40%市售药物作用于GPCR.然而,GPCR药物在中国水环境中赋存情况及生态风险还未知.基于药物行业生产报告构建GPCR药物环境浓度预测模型,使用体外测试检测22种高产量GPCR药物的生物活性,最后基于预测浓度和体外生物活性建立地表水中GPCR药物的环境活性预测模型,筛选出中国地表水中需要优先控制的13种GPCR药物. Pharmaceuticals with trace concentrations(ng/L-μg/L)have been widely detected in the aquatic environment.Concern about their potential ecological risks to aquatic species has been raised.G protein-coupled receptors(GPCR)are a class of membrane proteins that are highly conserved across species,and it is estimated that around 40%of marketed pharmaceuticals target GPCR.However,the occurrence and ecological risks of GPCR-acting pharmaceuticals in the aquatic environment in China remains unclear.Based on the pharmacy production report,an environmental concentration prediction model of GPCR-acting pharmaceuticals is developed.Then,the biological activities of 22 pharmaceuticals against corresponding GPCR with high production are investigated by an in vitro assay.Finally,a prediction model for environmental activities of GPCR-acting pharmaceuticals in surface water is established based on the predicted concentrations and in vitro biological activities,and 13 GPCR-acting pharmaceuticals are selected for priority control in surface water in China.
