特殊医学用途婴儿配方食品中叶黄素检测关键控制点研究

摘要 为了测定特医婴配食品中叶黄素含量,建立了高效液相色谱法,并对检测过程中关键控制点进行了详细研究。特医婴配食品样品经乙醇溶解分散,加入KOH溶液,于常温下进行皂化反应,经有机试剂提取浓缩后,用含0.1%BHT乙醇溶解定容,经C30色谱柱分离,紫外检测器检测,外标法定量。在优化的前处理及仪器分析条件下,特医婴配食品样品中的叶黄素含量测定在0.05~2μg/mL浓度范围内线性关系良好,相关系数为0.9999。高效液相色谱方法检出限(S/N=3)为1.5μg/100g,定量限(S/N=10)为5μg/100g。在5、50和250μg/100g添加水平下,高效液相色谱方法的平均回收率在85%~100%,相对标准偏差(RSD)为:0.89%~6.32%。方法对测定过程关键控制点的研究为特医婴配食品中不稳定成分叶黄素的准确测定奠定基础,为企业开展产品质量及原料控制提供了技术手段。 In order to determine the lutein content in special medical infant formula foods,an efficient liquid chromatography method was established,and detailed analysis was conducted on key control points in the detection process.Special medical infant formula food samples were dispersed in ethanol,treated with KOH solution for saponification at room temperature,extracted and concentrated with organic reagents,dissolved in ethanol containing 0.1%BHT for volumetric adjustment,separated by a C30 chromatographic column and detected by a UV detector,and quantification was performed using external standard method with HPLC.Under optimized pretreatment and instrumental analysis conditions,the lutein content in special medical infant formula food samples showed a good linearity within the concentration range of 0.05 to 2μg/mL,with a correlation coefficient of 0.9999.The detection limit of the high-performance liquid chromatography method(S/N=3)was 1.5μg/100g,and the quantification limit(S/N=10)was 5μg/100g.At the addition levels of 5,50,and 250μg/100g,the average recovery rates of the highperformance liquid chromatography method ranged from 85%to 100%,with relative standard deviations(RSD)of 0.89%to 6.32%.The study on key control points in the determination process provides a solid foundation for the accurate determination of the unstable component lutein in special medical infant formula foods and offers technical means for enterprises to carry out product quality and raw material control.
