摘要
为了确定福建省某商品肉鸡场出现的腿病病原,同时了解其病原的遗传进化特征和致病特点,对送检病鸡组织样品进行病原分离,并经PCR检测、测序分析等方法进行鉴定。结果:成功分离到1株呼肠孤病毒(ARV),命名为FJZQH23。该分离株在鸡肝癌细胞(LMH)上能够良好增殖,且有明显的细胞病变。ARV主要保护性抗原基因σC的遗传进化分析结果显示,分离株FJZQH23与Ⅰ型参考株4599隶属同一分支,两者亲缘关系较近。基于σC基因的氨基酸序列同源性分析发现,分离株FJZQH23与参考株4599相似度较高为86.9%,而与疫苗株S1133相似度为80.1%。对14日龄肉鸡致病性试验结果表明,分离株FJZQH23能够引起跗关节和脚垫出血。研究表明,基因Ⅰ型ARV感染为该肉鸡场的发病病原。本研究结果为该肉鸡场ARV感染的综合防控措施制定提供了依据。
In order to determine the pathogen of leg disease in a commercial broiler farm in Fujian Province,and at the same time to understand the genetic evolutionary features and pathogenic characteristics of the pathogen,this study was conducted to isolate the pathogen from the tissue samples of diseased chickens sent for examination,and a strain of avian reovirus(ARV)named FJZQH23 was successfully isolated,which was identified by PCR assay,sequencing analysis,etc.The results of genetic evolutionary analysis showed that the main protective antigen geneσC of ARV was able to proliferate well on LMH cells.The isolate was able to proliferate well on LMH cells with obvious cytopathic lesions.The genetic evolution analysis of σC,the main protective antigen gene of ARV,showed that the isolate FJZQH23 belonged to the same branch as the type I reference strain 4599,and the two strains were relatively close to each other.Based on the amino acid sequence homology analysis of theσC gene,the isolate FJZQH23 was 86.9%similar to the reference strain 4599 and 80.1%similar to the vaccine strain S1133.Pathogenicity tests on 14-day-old broilers showed that the isolate FJZQH23 was able to cause bleeding symptoms in the hock and foot pads.This study demonstrated that genotype I ARV infection was the pathogenic agent of the disease in this broiler farm,which provides a reference for the development of comprehensive preventive and control measures.
