摘要
目的:通过双向两样本孟德尔随机化(MR)方法探究疲乏与慢性疼痛之间的潜在因果关系。方法:在疲乏和慢性疼痛的全基因组关联研究(GWAS)汇总数据中分别提取显著相关且相互独立的变异位点作为工具变量。使用逆方差加权(IVW)、MR Egger回归、基于众数的简单估计(SM)、基于众数的加权估计(WM)和加权中位数(WME)进行双向两样本MR分析。结果:IVW方法显示疲乏频率的增加会显著增加慢性疼痛部位的数量(β=0.78,P<0.001),慢性疼痛部位数量的增加也会显著增加疲乏频率(β=0.35,P<0.001)。SM、WM和WME方法得出了相似的结果。结论:疲乏与慢性疼痛具有显著双向因果关系,应注意对该症状人群进行早期评估,及时干预。
Objective To explore the potential causal relationship between tiredness and chronic pain using bi-directional twosample Mendelian randomization(MR).Methods From the summary-level data of the genome-wide association study(GWAS)of tiredness and chronic pain,significant correlated and independent variation sites were extracted as instrumental variables.Inverse variance weighting(IVW),MR Egger regression,simple mode(SM),weighted mode(WM)and weighted median(WME)were used for the bi-directional two-sample MR analysis.Results IVW method showed that the increase of tiredness frequency significantly increased the number of chronic pain sites(β=0.78,P<0.001),and the increase of the number of chronic pain sites significantly increased the tiredness frequency(β=0.35,P<0.001).Similar results were obtained by SM,WM and WME methods.Conclusions There is a significant bi-directional causal relationship between tiredness and chronic pain,and attention should be paid to the early assessment and timely intervention in the population with these two symptoms.
