摘要
叶柄上叶基因(BLADE-ON-PETIOLE,BOP)参与调控植物生长发育中如叶片和花朵的形态形成和脱落等重要发育过程。本研究将蒺藜苜蓿R108作为材料,通过生物信息学分析、亚细胞定位及表达模式分析等实验,探究MtBOP基因的特性。MtBOP基因完整编码区为1452 bp,编码483个氨基酸。根据系统发育进化树结果可知其与豌豆(Pisum sativum)和红三叶草(Trifolium pratense)亲缘关系最近。通过信号肽预测分析,其不具有信号肽。亚细胞定位结果显示MtBOP定位于细胞核和细胞质。表达模式分析表明,该基因的表达量在脱落酸、细胞分裂素、水杨酸、赤霉素调节下有上调趋势,而在生长素,茉莉酸甲酯调节下出现下调趋势。酵母自激活检测显示MtBOP不具有自激活活性。本研究为进一步探究蒺藜苜蓿的MtBOP基因提供了科学依据,而且BOP可用于后续相关基因功能实验。
BLADE-ON-PETIOLE(BOP)gene is involved in regulating important plant developmental processes such as leaf and flower morphogenesis and shedding.In this study,Medicago truncatula R108 was used as a material to explore the characteristics of MtBOP through bioinformatics analysis,subcellular localization and expression pattern analysis.The complete coding region of MtBOP gene is 1452 bp,encoding 483 amino acids.According to phylogenetic tree results,it is closely related to Pisum sativum and Trifolium pratense.It has no signal peptide according to signal peptide prediction analysis.Subcellular localization results showed that MtBOP was located in the nucleus and cytoplasm.The analysis of expression pattern showed that the expression level of this gene was up-regulated under the regulation of abscisic acid,cytokinin,salicylic acid and gibberellin,while the expression of auxin and methyl jasmonate was down-regulated.The yeast self-activation test showed that MtBOP had no self-activation activity.This study provides scientific basis for further study of MtBOP gene of Medicago truncatula,and BOP can be used for subsequent related protein experiments.
