滇水金凤花蜜相关SWEET7和SWEET16的克隆及表达分析

摘要 植物SWEET(Sugars will eventually be exported transporter)基因家族是一类重要的糖转运蛋白,参与开花植物的花蜜合成。本研究以野生型和突变型滇水金凤(Impatiens uliginosa)为材料,基于课题组前期的花距转录组数据,通过筛选和RT-PCR技术克隆得到花蜜相关基因SWEET7和SWEET16,分别命名为IuSWEET7和IuSWEET16,其cDNA分别为741 bp和903 bp,分别编码246和300个氨基酸。生物信息学分析表明:IuSWEET7为疏水性不稳定蛋白,IuSWEET16为疏水性稳定蛋白,二者均含有2个典型的MtN3/saliv保守结构域;IuSWEET7和IuSWEET16基因的氨基酸序列与杜鹃花(KAG5539487.1)、一串红(XP_042052415.1)等植物同源序列的相似性均在54.15%~71.48%;系统进化分析表明,IuSWEET7和IuSWEET16处于两个不同分支。qRT-PCR分析表明两个基因在野生型和突变型滇水金凤花距的3个时期中均有表达,且在不同部位中表达模式不同。其中IuSWEET7基因在野生型滇水金凤中其表达量从花苞期至盛花期逐渐上升;在突变型2距和3距中其表达量从花苞期至盛花期先上升后下降,且在始花期表达量最高;而IuSWEET16基因在野生型和突变型3距中其表达量从花苞期至盛花期均逐渐上升,但在突变型2距中其表达量从花苞期至盛花期先上升后下降,也在始花期时达到最高。 The SWEET(Sugars will eventually be exported transporter)gene family in plants is an important class of sugar transporter proteins involved in nectar synthesis in flowering plants.In this study,wild type and mutant Impatiens uliginosa were used as materials,the transcriptome data of the flower spur generated by our research group were used,we identified and cloned the nectar-related genes SWEET7 and SWEET16 through screening and RT-PCR techniques,designating them as IuSWEET7 and IuSWEET16,respectively.Their cDNA were 741 bp and 903 bp,encoding 246 and 300 amino acids,respectively.Bioinformatics analysis showed that IuSWEET7 was a hydrophobically unstable protein,while IuSWEET16 was a hydrophobically stable protein,both containing two typical MtN3/saliv conserved domains;the amino acid sequences of IuSWEET7 and IuSWEET16 genes showed similarity ranging from 54.15% to 71.48% to the homologous sequences in Rhododendron griersonianum and Salvia splendens;phylogenetic analysis showed that IuSWEET7 and IuSWEET16 belonged to two different branches.QRT-PCR analysis showed that both genes were expressed across three stages of flower spur in both wild-type and mutant I.uliginosa,with varying expression patterns in different organs.The expression of IuSWEET7 gene in wild-type I.uliginosa exhibited a gradual increase from the bud stage to the flowering stage.In the mutants,the expression first increased and then decreased in 2 and 3 spurs of mutants type,with the highest expression observed during the first flowering stage.The expression of IuSWEET16 gene increased gradually from the bud stage to the flowering stage in wild-type and 3 spur of mutant type.However,in the second spur of the mutant type,the expression increased at first and then decreased from the bud stage to the flowering stage,reaching its peak at the first flowering stage.
