摘要
目的优化新疆藁本总黄酮的的提取工艺,并评价其体外抗氧化及降糖活性。方法采用单因素及响应面法优化新疆藁本总黄酮的提取工艺,用HPLC法测定主要黄酮苷元的分布及含量。以DPPH自由基、羟基自由基、ABTS自由基的清除率来评价新疆藁本总黄酮的抗氧化能力;采用α-葡萄糖苷酶抑制实验评价其体外降糖能力。结果超声辅助提取新疆藁本总黄酮的提取量最高,优化后新疆藁本总黄酮的提取量为21.36 mg·g^(-1)。从新疆藁本总黄酮提取液中共鉴定出6个主要黄酮类化合物,其中含量较高的是葛根素、木犀草苷、槲皮苷,分别为6.972、2.921、3.257 mg·g^(-1)。新疆藁本总黄酮对DPPH自由基、羟基自由基、ABTS自由基均具有优异的清除能力,并在一定的浓度范围内呈现出剂量依赖性;当新疆藁本总黄酮为1.4846 mg·mL^(-1)时,对α-葡萄糖苷酶的抑制率可达91.23%。结论优化后的新疆藁本总黄酮提取方法,含量测定方法准确性高,可用于提取和预测新疆藁本总黄酮的提取量。
OBJECTIVE To optimize the extraction process of total flavonoids from Conioselinum tataricum Hoffmann,and to evaluate their in vitro antioxidant and hypoglycemic activities.METHODS Single factor and response surface experiments were used to optimize the extraction process of total flavonoids from C.tataricum.The distribution and content of the main flavonoid aglycones were identified by HPLC.The antioxidant capacity of total flavonoids from C.tataricum was evaluated by DPPH free radical scavenging rate,hydroxyl radical scavenging rate,and ABTS free radical scavenging rate.The in vitro hypoglycemic activity was evaluated by theα-glucosidase inhibition experiment.RESULTS After optimization,the total flavonoids extracted from C.tataricum was 21.36 mg·g^(-1).Six major flavonoids were identified from the total flavonoid extracts.Among them,the higher contents were puerarin,luteolin,and quercetin,which were 6.972,2.921,and 3.257 mg·g^(-1),respectively.All of them showed excellent scavenging capabilities and a dose dependence within a certain concentration range.When the total flavonoid mass concentration of C.tataricum was 1.4846 mg·mL^(-1),theα-glucosidase inhibition rate reached 91.23%.CONCLUSION The established method for extracting total flavonoids from C.tataricum has high accuracy and reliability,and can be used to extract and predict the amount of total flavonoids extracted from C.tataricum.
