二甲双胍对D-半乳糖诱导的衰老小鼠肝脏组织的影响

摘要 旨在研究二甲双胍对衰老小鼠肝脏组织的影响.诱导灌胃结束后处死小鼠,称其体质量和肝脏质量,HE染色观察肝脏形态学变化,检测小鼠肝脏组织中活性氧(ROS)、总抗氧化能力(T-AOC)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及还原型谷胱甘肽(GSH)、丙二醛(MDA)浓度,同时检测衰老分泌表型相关基因P53,P21,P16,LaminB1,HMGB1,IL-6,TNF-α,CXCL-10,Rad51的表达变化.结果显示,与对照组(Ctrl)相比,诱导衰老组(IA)ROS,CAT活性和MDA浓度均显著升高,T-AOC,SOD,GSP-Px活性和GSH浓度均显著降低,衰老分泌表型相关基因mRNA表达水平均升高.与IA组相比,在诱导衰老+二甲双胍组(IA+Metf)中,ROS,CAT活性和MDA浓度均显著降低,T-AOC,SOD,GSP-Px活性和GSH浓度均升高,小鼠肝脏衰老分泌表型相关基因mRNA表达水平均降低.结果表明,二甲双胍对D-半乳糖诱导的衰老模型小鼠肝脏损伤具有改善作用,可为二甲双胍在抗衰老中的利用提供实验依据. The purpose is to investigate the impact of metformin on liver tissue in aging mice.After induction gavage,the mice were euthanized,and their body quality and liver quality were measured.HE staining was utilized to observe morphological changes in the liver tissue.Additionally,activity of reactive oxygen species(ROS),total antioxidant capacity(T-AOC),catalase(CAT),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and the concentrations of reduced glutathione(GSH),malondialdehyde(MDA)in mouse liver tissue.Simultaneously,the expression levels of genes related to aging secretion phenotype including P53,P21,P16,LaminB1,HMGB1,IL-6,TNF-α,CXCL-10 and Rad51 were also detected.The results revealed that compared with the control group(Ctrl),the ROS,CAT activity,and MDA concentration in the induced aging group(IA)were significantly increased,while the T-AOC,SOD,GSP-Px activity and GSH content were significantly reduced.Additionally,the mRNA expression levels of genes related to aging secretion phenotype were all elevated.Compared with the IA group,in the induced aging+metformin group(IA+Metf),ROSlevels,CAT activity,and MDA concentration were significantly reduced,while the activities of T-AOC,SOD,GSP-Px and GSH contents increased.Further more,the mRNA expression levels of genes related to aging secretion phenotypein mouse liver are reduced.In summary,metformin has an improving effect on liver damage in D-galactose-induced aging model mice,which can provide experimental evidence for the utilization of metformin in anti-aging.
