摘要
目的:探索血浆脂质体与双向情感障碍(BD)间的因果关联。方法:从全基因组关联研究(GWAS)的汇总统计数据库中获取179种血浆脂质体和BD的GWAS数据,将与血浆脂质体有关的单核苷酸多态性(SNPs)作为工具变量用于初步双样本孟德尔随机化(MR)分析。对初步MR分析中符合筛选要求的脂质体进行复制分析与mate分析,以进一步探索脂质体与BD间因果关联的稳健性。结果:初步MR分析显示有13种血浆脂质体与BD存在因果关联,这种因果关联经FDR校正后消失。但mate分析结果显示,磷脂酰胆碱(16:0_18:1)(OR=1.123,P=0.001)、磷脂酰胆碱(16:0_18:3)(OR=1.087,P=0.001)、磷脂酰胆碱(16:0_20:2)(OR=1.092,P=0.001)、三酰基甘油(52:5)(OR=1.090,P=0.001)、三酰基甘油(51:3)(OR=1.081,P=0.001)、三酰基甘油(52:3)(OR=1.052,P=0.023)、鞘磷脂(d36:1)(OR=1.046,P=0.031)是BD的潜在危险因素,而磷脂酰乙醇胺(O-16:1_22:5)(OR=0.931,P=0.004)是BD的保护因素。结论:血浆脂质体与BD存在潜在的因果关联,但具体的作用机制还需实验性研究进一步论证。
Objective:To investigate the causal relationship between the plasma lipidome and bipolar disorder(BD).Methods:We obtained the genome-wide association study(GWAS)data for 179 plasma lipidomes and BD from publicly available summary statistical databases and set single-nucleotide polymorphisms(SNPs)related to the plasma lipidomes as instrumental variables for an initial two-sample Mendelian randomization(MR)analysis.We conducted replication analysis and mate-analysis on the lipidomes that meet the selection criteria in the preliminary MR analysis to further explore the robustness of the causal association between lipidome and BD.Results:The preliminary MR analysis showed a causal association between 13 kinds of plasma lipidomes and BD,which disappeared after FDR correction.However,the mate analysis results showed that phosphatidylcholine(16∶0_18∶1)(OR=1.123,P=0.001),phosphatidylcholine(16∶0_18∶3)(OR=1.087,P=0.001),phosphatidylcholine(16∶0_20∶2)(OR=1.092,P=0.001),triglyceride(52∶5)(OR=1.090,P=0.001),triglyceride(51∶3)(OR=1.081,P=0.001),triglyceride(52∶3)(OR=1.052,P=0.023),and sphingomyelin(d36∶1)(OR=1.046,P=0.031)were potential risk factors for BD,while phosphatidylethanolamine(O-16∶1_22∶5)(OR=0.931,P=0.004)is a protective factor for BD.Conclusion:The potential causal association between the plasma lipidome and BD,but the specific mechanisms still need further experimental research to validate.
