湖南省怀化市鸡蛔虫流行情况调查及其遗传多样性与种系发育关系研究

A survey on the prevalence of Ascaridia galli in Huaihua City,Hunan Province,and study on the relationship between its genetic diversity and phylogenetic development

摘要 为调查湖南省怀化市鸡蛔虫的流行情况,探究其遗传多样性,阐述其种系发育关系,试验以采自湖南省怀化市不同地区的430份鸡粪便,用饱和盐水漂浮法检测虫卵,利用IBM SPSS Statistics 22软件统计、分析数据,并用χ^(2)法检验各地区、各类型鸡感染鸡蛔虫的差异性;PCR扩增其cox1与nad4基因的部分序列并测序;运用Clustal X、DNAStar软件分析基因的碱基组成与遗传多样性;运用Clustal X、PhyML 3.0软件分析鸡蛔虫的种系发育关系。结果表明:湖南省怀化市鸡蛔虫的总感染率为56.98%,其中圈养鸡的感染率为9.07%,放养鸡的感染率为47.91%,各地区、各类型鸡对鸡蛔虫的感染存在一定程度的差异;鸡蛔虫cox1基因序列的长度为953 bp,A、T、G、C碱基含量分别为19.0%~19.3%、45.9%~46.5%、23.0%~23.2%、11.3%~12.0%,A+T及G+C含量分别为64.9%~65.8%和34.3%~35.2%,种内差异为0~2.1%,种间差异为13.12%~14.48%;nad4基因序列的长度为876 bp,A、T、G、C碱基含量分别为19.1%~19.4%、52.4%~52.9%、19.9%~20.2%、7.9%~8.3%,A+T及G+C含量分别为71.5%~72.3%和27.8%~28.5%,种内差异为0~2.0%,种间差异为26.37%~27.40%;来自于湖南省怀化市的鸡蛔虫均处于发育树的一个分支上。说明湖南省怀化市鸡蛔虫的流行情况较严重,其cox1、nad4基因序列均呈AT偏好,均具有种内差异小而种间差异大的特征,是一类良好的遗传标记,且其个体之间虽存在一定程度的基因变异和遗传多样性,但具有较近的亲缘关系。 In order to investigate the prevalence of Ascaridia galli in Huaihua City,Hunan Province,and to explore its genetic diversity and to elaborate the relationship of its phylogenetic development,in the experiment,a total of 430 chicken manure samples were collected from different areas of Huaihua City,Hunan Province,and the eggs were detected by saturated brine floating.IBM SPSS Statistics 22 software was used to collect and analyze the data,and the chi-square method was used to test the differences of Ascaridia galli infection in different areas and types of chicken.Some sequences of cox1 and nad4 genes were amplified and sequenced by PCR;the base composition and genetic diversity of the two genes were analyzed by Clustal X and DNASTAR software,and the phylogenetic relationship between Ascaridia galli was analyzed by Clustal X and PhyML 3.0 software.The results showed that the total infection rate of Ascaridia galli in Huaihua area of Hunan Province was 56.98%,including 9.07%in captive chickens and 47.91%in free-range chickens.There were some differences in the infection of Ascaridia galli in different areas and types of chickens.The length of the cox1 gene sequence was 953 bp,and the contents of four bases A,T,G,and C were 19.0%-19.3%,45.9%-46.5%,23.0%-23.2%,and 11.3%-12.0%,respectively;and the contents of A+T and G+C were 64.9%-65.8%and 34.3%-35.2%,respectively;and the intraspecific differences were 0-2.1%,and the interspecific differences were 13.12%-14.48%,respectively.The length of nad4 gene sequence was 876 bp,and the contents of four bases A,T,G,and C were 19.1%-19.4%,52.4%-52.9%,19.9%-20.2%,and 7.9%-8.3%,respectively;the contents of A+T and G+C were 71.5%-72.3%and 27.8%-28.5%,respectively;the intraspecific differences were 0-2.0%,and the interspecific differences were 26.37%-27.40%,respectively.Ascaridia galli from Huaihua area of Hunan Province was located on a branch of the phylogenetic tree.The results indicated that the epidemic of Ascaridia galli was serious in Huaihua City,Hunan Province,and the cox1
