陕西省榆林地区犬弓首蛔虫的流行情况调查及其遗传多样性与种系发育关系分析

Epidemiological survey of Toxocascaris canis in Yulin area of Shaanxi Province and analysis of the relationship between genetic diversity and germline development

摘要 为了调查陕西省榆林地区犬弓首蛔虫的流行情况,并研究其基因变异、遗传多样性及种系发育关系,试验采用饱和盐水漂浮法对采自该地区的402份犬粪便进行虫卵检查,利用IBM SPSS Statistics 22软件对虫卵检测数据进行统计和分析,并采用χ^(2)检验分析各地区、各类型犬感染犬弓首蛔虫的差异性;PCR扩增犬弓首蛔虫的cox1、18S rRNA基因序列并测序;运用Clustal X与DNAStar软件分析2个基因的碱基组成与遗传多样性;运用Clustal X及PhyML 3.0软件分析犬弓首蛔虫的种系发育关系。结果表明:陕西省榆林地区犬对弓首蛔虫的总感染率为20.65%,其中宠物犬的感染率为8.42%,而流浪犬的感染率为33.00%,各地区、各类型犬感染犬弓首蛔虫的情况存在一定差异;cox1基因序列长度为1044 bp,A+T含量为61.5%~62.2%,G+C含量为37.8%~38.5%,种内差异为0~1.3%,种间差异为9.87%~14.37%;18S rRNA基因序列长度为598 bp,A+T含量为50.0%~50.5%,G+C含量为49.5%~50.0%,种内差异为0~1.4%,种间差异为0.33%~6.68%。在2个种系发育树中,犬弓首蛔虫均互相掺杂在一起共同形成每个种系发育树的一个分支。说明犬弓首蛔虫在榆林地区的流行情况较为严重,各个体之间虽存在较低程度的基因变异和遗传多样性,但具有较近的亲缘关系。 In order to investigate the prevalence of Toxocara canis in Yulin,Shaanxi Province,and study the relationship between gene variation,genetic diversity and germline development,a total of 402 dog feces collected from this area were examined for their eggs by the saturated salt water floating method.The software IBM SPSS Statistics 22 was used to make statistics and analysis on the egg detection data,andχ^(2)test was used to analyze the difference of Toxocara canis infection rate among different regions and types of dogs.The cox1 and 18S rRNA gene sequences of Toxocara canis were amplified by PCR and sequenced.The software Cluster X and DNAStar were used to analyze the base composition and genetic diversity of the two genes.The software Cluster X and PhyML 3.0 were used to analyze the phylogenetic relationship of Toxocara canis.The results showed that the total infection rate of dogs was 20.65%;the infection rate of pet dogs was 8.42%,and the infection rate of stray dogs was 33.0%.There were some differences between different regions and different types of dogs.The length of cox1 gene sequence was 1044 bp;the content of A+T was 61.5%-62.2%;the content of G+C was 37.8%-38.5%;the differences within the species were 0-1.3%,and the differences between the species were 9.87%-14.37%.The sequence length of 18S rRNA was 598 bp,the content of A+T was 50.0%-50.5%;the content of G+C was 49.5%-50.0%,and the differences between species were 0-1.4%and 0.33%-6.68%.In both germline developmental trees,Toxocara canis was intermixed to form a branch of each germline developmental tree.These results indicated that the prevalence of Toxocarria canis was serious in Yulin area.Although there was a low degree of genetic variation and genetic diversity among individuals,there was a close genetic relationship.
