摘要
本研究旨在评估CpG和poly I:C对口蹄疫(FMD)病毒样颗粒(VLPs)疫苗的免疫增强效果。以纯化的O型FMDVLPs抗原为基础,使用VLPs-CpG和VLPs-poly I:C刺激小鼠骨髓源性树突状细胞(BMDC),通过流式细胞术分析CD40和CD86在BMDC中的表达水平。进一步将VLPs、VLPs-CpG和VLPs-poly I:C分别与ISA206乳化制备疫苗,并免疫豚鼠,通过检测特异性抗体、中和抗体、脾淋巴细胞增殖以及攻毒试验评估CpG和poly I:C对VLPs疫苗免疫效果的影响。结果显示,VLPs-CpG组和VLPs-poly I:C组的CD86表达水平高于VLPs组,ISA206佐剂联合poly I:C、CpG均能提高攻毒保护率,而且ISA206联合CpG可以加速特异性抗体及中和抗体反应,提高脾淋巴细胞增殖水平。本研究结果表明,CpG和poly I:C均能增强FMDVLPs疫苗的免疫效果,具有潜在的开发应用价值。
This study was to evaluate the immune-enhancing effects of Cp G and poly I:C on the footand-mouth disease(FMD)virus-like particles(VLPs)vaccine.Using purified serotype O FMD VLPs as antigens,the mice bone marrow-derived dendritic cells(BMDC)were stimulated with VLPs-CpG and VLPs-poly I:C followed by the expression levels of CD40 and CD86 in BMDC was analyzed using flow cytometry.Further,VLPs,VLPs-Cp G and VLPs-poly I:C were emulsified with ISA206 to immunize guinea pigs.The improvement of Cp G and poly I:C on immune effect of the VLPs vaccine was evaluated by detecting the specific antibodies,neutralizing antibodies and splenic lymphocyte proliferation,as well as virus challenge.Results showed that the expression levels of CD86 in VLPs-Cp G and VLPs-poly I:C were higher than VLPs.The adjuvant ISA206 combined with poly I:C and Cp G could improve the protection against virus.ISA206 combined with Cp G accelerated the responses of specific antibodies and neutralizing antibodies,as well as enhanced the proliferation of splenic lymphocytes.This study demonstrates both Cp G and poly I:C has application potential to enhance the immune effect of FMD VLPs vaccine.
