二重实时荧光RT-PCR检测法在口蹄疫病毒检测中的应用

摘要 为探究二重实时荧光RT-PCR检测法在口蹄疫病毒检测中的应用情况,依照口蹄疫病毒(FMDV)聚合酶3D基因序列,规划并组合成引物与探针,有针对性的优化部分反应条件,构建了FMDV通用型与A型二重实时荧光RT-PCR检测法。选择124份疑似感染的试样进行检测。结果显示,构建的荧光RT-PCR方法在101~107拷贝/uL模板范围中形成良好的线性关系,对pGEM-T/FMDV-T和pGEM-T/FMDV-A重组质粒均呈现出阳性扩增信号,共计检测出FMDV-T阳性10份,FMDV-T/FMDV-A双阳性7份。可见,RT-PCR检测法能够定量分析及同时检测大量口蹄疫病毒样本。
