白桦脂酸及其衍生物的抗肿瘤作用和机理研究进展

摘要 白桦脂酸(Betulinic acid,BA)为羽扇豆烷型五环三萜类化合物,在马甲子、杜仲和酸枣仁等多种中药中均含量较高。既有研究显示白桦脂酸具有抗肿瘤、抗炎、抗HIV、抗疟疾、抗菌和抗氧化等多种生物活性,尤其是在抗肿瘤方面,因其低毒、高效的特点而受到广泛重视,相关机理研究也较多,包括线粒体途径、Sp转录因子途径以及自噬等,但总体并不清晰。本文对白桦脂酸抗肿瘤作用机制进行综述,分瘤种厘清现有发现,分析研究热点,以期为白桦脂酸和以白桦脂酸及其类似物为主要药效成分的抗肿瘤药物研究提供参考。 Betulinic acid(BA)is a lupinane-type pentacyclic triterpenoid with high content in a variety of Chinese medicinal materials,such as Majiazi(马甲子),Duzhong(杜仲),and Suanzaoren(酸枣仁).Studies have demonstrated that BA has anti-tumor,anti-inflammatory,anti-HIV,anti-malarial,antimicrobial,and antioxidant activities.It has attracted much attention in the treatment of tumors because of the low toxicity and high efficiency.Accordingly,extensive studies have been conducted about the related mechanisms,including the mitochondrial pathway,the Sp transcription factor pathway,and autophagy,while the overall picture remains unclear.In this paper,we reviewed the anti-tumor mechanism of BA,clarified the existing findings regarding different tumors,and analyzed the research hotspots,aiming to provide references for the research on BA and anti-tumor drugs with BA and its analogs as the main pharmacodynamic components.
