摘要
目的:分析蒲公英甾醇对口腔鳞癌SCC-9细胞增殖、凋亡的影响,并探究其潜在机制。方法:采用不同浓度蒲公英甾醇处理人口腔鳞癌SCC-9细胞。通过CCK-8实验分析不同浓度蒲公英甾醇对SCC-9细胞增殖的影响;通过蛋白免疫印迹实验检测不同浓度蒲公英甾醇处理后SCC-9细胞中p53蛋白和增殖、凋亡相关蛋白的相对表达量;通过RT-qPCR检测p53 mRNA的表达。结果:相较于对照组,不同浓度蒲公英甾醇显著抑制SCC-9细胞的增殖(P<0.05),抑制作用与蒲公英甾醇的浓度与作用时间成正比;蒲公英甾醇显著促进SCC-9细胞凋亡(P<0.05),并存在剂量效应作用;蒲公英甾醇显著促进SCC-9细胞p53 mRNA的表达以及p53蛋白的表达(P<0.05)。结论:蒲公英甾醇激活p53信号通路,抑制口腔鳞状细胞癌SCC-9细胞增殖,促进SCC-9细胞凋亡。
Objective:To investigate the effect of taraxasterol on the proliferation,apoptosis,and p53 expression of oral squamous cell carcinoma SCC-9 cells,and to explore the potential mechanism.Methods:Human oral squamous cell carcinoma SCC-9 cells were treated with various concentrations of taraxosterol.The CCK-8 assay was utilized to examine the impact of different concentrations of taraxasterol on the proliferation of SCC-9 cells.Western blot assay was used to detect the effect of taraxasterol on the expression of p53,proliferation,and apoptosis-related proteins in SCC-9 cells.The expression of p53 mRNA was detected using RT-qPCR.Results:Compared with the control group,different concentrations of taraxasterol could signally inhibit the proliferation of SCC-9 cells(P<0.05).The inhibitory effect was directly proportional to the concentration and duration of exposure to taraxasterol.Taraxasterol promoted the apoptosis of SCC-9 cells(P<0.05)in a dose-dependent manner.Taraxosterol can obviously enhance the expression of p53 mRNA and p53 protein in SCC-9 cells(P<0.05).Conclusion:Taraxasterol inhibits the proliferation and promotes the apoptosis of oral squamous cell carcinoma SCC-9 cells by regulating the p53 signaling pathway.
