高效液相色谱法测定化妆品中白藜芦醇

摘要 建立高效液相色谱法测定化妆品中白藜芦醇,并用质谱确证。样品经甲醇涡旋超声提取、离心后,通过Merck Discovery■HS C_(18)(250 mm×4.6 mm,5μm)色谱柱,采用甲醇-0.05%磷酸水溶液(体积比为40∶60)为流动相,等度洗脱,流量为1.0 mL/min,柱温为30℃,检测波长306 nm,进样量为10μL,采用负离子多反应监测模式进行质谱确证。白藜芦醇在质量浓度1.0~100μg/mL范围内与色谱峰面积线性关系良好,相关系数为0.9998,检出限和定量限分别为3.0和10.0 mg/kg,在膏霜、油和水三种不同基质中测定值的相对标准偏差均小于5.0%(n=10),加标平均回收率为87.1%~96.6%。该方法样品处理简便,稳定性较好,定量结果准确,适用于化妆品中白藜芦醇的高效测定。 A high-performance liquid chromatography method was developed for the determination of resveratrol in cosmetics and confirmed by mass spectrometry.The procedure involved extraction of samples through methanol vortex sonication and centrifugation,followed by separation on a Merck Discovery®HS C_(18) column(250 mm×4.6 mm,5μm)utilizing isocratic elution with methanol-0.05%aqueous phosphoric acid(40:60,v/v)as the mobile phase.The flow rate was 1.0 mL/min,with a column temperature of 30℃.The detection wavelength was 306 nm,and the injection volume was 10μL.Mass spectrometric confirmation was conducted in negative ion multiple reaction monitoring mode.The mass concentration of resveratrol had a good linear relationship with the chromatographic peak area in the range of 1.0-100μg/mL,and the correlation coefficient was 0.9998.The detection and quantification limits were 3.0 mg/kg and 10.0 mg/kg,respectively.The relative standard deviations of values obtained from three different matrices,such as creams,oils,and water,were all below 5.0%(n=10).The average recovery of standard addition was 87.1%-96.6%.The method has the advantages of simple sample preparation,good stability and accurate quantitative results,which is suitable for the efficient determination of resveratrol in cosmetics.
